Been spamming these a lot lately but here you go, tampanensis bottle grow on rye with gypsum:

Cased with potting soil.

I'll have stuff like Pan. cyan and Ps. mexicana to show soon too.

tampanensis:

After germinating the spores on pda agar, I made transfers to new plates. These new plates pinned and produced fruits before the culture was even half the size of the pasty plate.

so I cloned one of those fruits and after a few transfers was able to grow a plate all the way out without it pinning out. So I made some grain spawn. I used this grow to experiment with my now published blenderless liquid inoculate tek. the culture was flat and solid white

1qt spawn, 1qt sub inside a 6 qt. sterlite shoebox with the lid on. after 100%, misted almost daily

outgrew their home, converted to dubtubs

harvest

yield: ±1/3oz. dry per tub (1qt spawn/1qt sub)
potency: very potent, close to PE
taste: metallic and horrendous bitter aftertaste

notes: the fruits are very thin and high swings of rh can easily dry them out. make sure you have a properly dialed in FC method for these. I had to mist them daily to prevent them from drying out. the dubtub was pretty high fae so i had to pay close attention to them.

I also have a Tamp project in the works...

I'm down to four jars now, one had mold...One of the jars appears to have begun forming sclerotia before colonizing 100%. These jars will be for stones....The tub is a mix of Hpoo, straw, verm, with one quart spawned, just looking for fruits off this tub...

I have a 2nd project almost ready...10 colonized agar dishes to 10 jars of rye...plan is 5 more jars for stones, 5 more to spawn for fruits...

Got a few plates of pan cyans on mea and 4 quart jars of wbs noc'd up with a little bit of agar if that qualifies for this thread. The pan myc is moving slow on agar. Very sluggish compared to cubes. I had a hunch that it would begin to take off once it hit grain.

So since i was beginning to lose patience i took a few of my plates with the most growth (only had myc growth about the size of a quarter) and noc'd up 4 jars. Ideally i would have waited til i had more substantial myc growth before transferring to grain.

The jars are less than 48hours since inoculation and already im seeing more growth than i would have on agar in that amount of time.

Also i used a print on agar to start with like i typically do with cubes. Im beginning to think that making a spore syringe first to noc up the agar instead, would have been more advantageous. Giving the spores a chance to rehydrate and what not before going to agar. Maybe would've aided in germination which seemed to take forever. What do y'all think about that subject?

I find i almost never make spore syringes anymore or lc for that matter. Havent made any lc in ages

Psilocybe allenii --> spore print --> syringe --> modified pftek. inoculated 11/27. First timer looking good so far. Neither my phone or my old digital camera can capture a close up/macro shot but i assure you this is healthy mycelium not cobweb or mold. Just starting to go from cottony to rhizomorph for lack of vocabulary.i hope this is considered exotic. If not my bad. I found 4 specimens this year near seattle in my 10 years of hunting so they are exotic to me, thats the only reason im attempting to cultivate actually.

It's most deff exotic. I'm doing cyanescens in a similar way. I made a slurry from pf tek jars and inoculated a couple 7 gal pots of wood chips n top soil the other day. I hope they're fine after the snow we had the other day.

Yes rye berries.  I started by cloning a wild fruit to agar.  Found the patch about 2 blocks from my house so I know it will do well in my backyard.

Out of 10 pastyplates 8 or so contaminated, but the 2 that didn't were expanded to 16 more.  From there I dropped dime sized agar wedges in rye grain jars.  With such a small piece of agar it's taking a little while to colonize.  It also doesn't help that my house temp is 65.

Hey amidogen, TMC says stones form best when grown in total darkness...might help if you don't already have them in the dark.  Also, looks like folks have had great success adding sand to the casings of their tamps or other stone producers...

Edit: here's one such grow:

2 Jars of WBS I knocked up via Ps. cyanescens MS syringe on 11/19/2016.
Yes I know, :dumblol:, I didn't have agar at the time but it finally came in yesterday........:bearbreakdance:
This was mainly for practice but I also have a bag of 3 ml, 25 G syringes a buddy gave me which at first I rendered pretty much useless but I was curious to see if I could end up using these if I was ever in a pinch.
I used 1 syringe and a tiny bit of spores for the 2 jars, so 1.5 ml each jar.

11/28/2016

12/14/2016

I shook them yesterday.

Also putting some Ps. semilanceata spores on agar tonight.

Next time I would use far far far less spore solution. A singular drop is all that's needed.

One disadvantage of bags is if you put in a lot of grain you won't find much sclerotia in the center of the bag. Usually sclerotia is forming between the bag/glass and the grains.

That's why I like to use long jars like this for sclerotia producing species:

~ 85x210mm

That's a sweet jar.

Why do they generally form on the sides of bags/glass.
Seems like it would be through the entire thing.

I've never worked with stones so idk the science behind it.

I often noticed that the center does not produce sclerotia.
I am working on an isolate (Psi. galindoi) right now to test if the production of sclerotia can be stimulated by adding marbles to the substrate to increase the surface area. I want to do this with an isolate to get legit test results.

An interesting post of a pretty unknown user:

Quote:

There is PLENTY of undigested grain in the middle of the jar (colonized, but still visible grain with visible starch etc), so I don't Beleive any low yield has much to do with lack of nutes... It's strange too because the way I understand sclerotia and their biological function is that they form in grasslands a the dry season approaches, to protect against drough. So sclerotia should form under drier conditions, yet here they are forming in the highest humidity against te glass with condensation droplets around and such, yet the middle of the jar with undigested grain is actually pretty dry... Why? What's happening here?
Here's my thoughts: sclerotia Indeed form in response to drought conditions, but rather than waste energy producing modifications everywhere within their mycellial network, they focus this energy in area where they meet physical constraints/resistance, something hard that it can't easily digest.. In nature this would be a fat tree root or rock. Now, when the rain finally comes again, if you clumped up your sclerotia bodies around large hard surfaces, I would think that the rainwater would collect around and follow that hard surface according to gravity. Kinda like rain collecting and streaming off the sides of your moving car, even tho the rain isn't coming down hard at all, maybe just a drizzle. Evolved habits to help guarantee rehydration during wet season... Just my thoughts, maye I'm way off...

I shortened the quote a bit.

Another great idea, Th3Issu3.
I open a new thread when I completed the agar work.

In that post a shot glass was a suggestion. Good ideas

Enlightenment

Have you tried this on an ms? Or have you been working out a proven isolate ?
It almost seems as if they form from empty spaces between grains next to the glass. Maybe larger grains could be used to achieve the larger spacing (more stones).

So true, great ideas guys, holy shit this gave me a wild idea of custom making a slim glass/plastic container (Like a window but with a 3 or 5 cm wide space in the middle).

And just throwing in colonised grains inside, and make a sort of trap door at the bottom which can slide off which you can just drop all the grains when you want to harvest.

Hang it like a picture on the wall, its literally a growing and live art show

Stonesun let his bags colonize not full and flat so there was more bag surface area per grain for the stones to grow.

GTK!  Makes sense as the stones would be just sub-surface as opposed to 6 inches down....I would imagine.

Giving the spores a chance to re-hydrate and what not before going to agar. Maybe would've aided in germination which seemed to take forever. What do y'all think about that subject?

I find i almost never make spore syringes anymore or lc for that matter. Havent made any lc in ages

So my hunch that the pan cyan myce would take off once it hit grain was on point. Something about the MEA, the pans apparently didn't care for it. Really sluggish growth.

A quick search and you'll find numerous posts with people using malt extract for their pans.  It does indeed work but im thinking there's something more preferable to use.

Note the growth on each of these side by side pics.

The pic on the left is from 11 days and the pic on the right is the dish today.  As you can see it's only doubled in size in that time.  In that amount of time it would've grown all the way to the end of the dish and began crawling up the sides looking for nutrients if it was p.cubes.

Now scope the jars out.  I shook em I wanna say like 5ish days ago, now they're about done.  I didn't have a whole hell of a lot of pan myce on agar initially so I only ended up noc'n up 3 quart jars(which are all looking nearly exactly the same now).  So about 15 days for full colonization of a quart jar using just a small piece of myce from the agar dish, not too bad.

Check out this nifty little alcohol torch I found.

You just give it a little squeeze and you can direct the flame.  Gets a scalpel red hot quite quickly.  I really like this little torch, so convenient.  A lot of times I dont even have to squeeze it.  I just let is sit on the table, light it up real quick with a lighter, hold the scalpel above the flame to heat it up and just blow it out when done.

So I set 2 quart jars aside for a mini grow with the pan cyan spawn I had. The rest went to g2g.

I did cvg + manure for the Pan, no casing. Got some scraggly looking pins
.. we'll  see.
