Spores, liquid culture or spawn. And to what substrate, and how was the substrate prepared. Any pictures?

oyster were from dowels to grain to those bags of grain. and king oyster were from bought grain to those bags of grain.
the grain media is wheat grain soaked for 18 hours with coffee, autoclaved for 2 hours to 15 psi.

I think the coffee has made it too acidic, grains are already acidic. Skip the coffee, use that with wood in a substrate.

As I said the problem is you added coffee to the grain, making it too acidic. If a substrate is too acidic or to alkaline it will grow very slow.
Stalling and then damage if too acidic to start with.

Jeez guys... Patience!! Don't start worrying for a week at least. The KO will be slower.
Though as Ferather said, skip the coffee and any other voodoo juice for grain (and IMO sub) - though I seriously doubt it has had a negative effect.

The dowels will take 2 weeks shake around day 7-8 . If you don't put dowels next to glass they will appear to do nothing. patience

Just want to start a little mushroom business. So I just want to have the best strain for boosting profit. So running late is not an option. I'm reading a lot on this and want to try strain isolation. Can't find a lot of informations on this particular subjet.
I will cut the coffee in my next grain spawn. I own a ph pen... didn't check the ph after soaking the grain
Another question I have: when making water culture when could you tranfer the water culture to grain? Do I need to wait for the spawn to inoculate the water or can I use it directly? Thanks!

If you purchased lc it probably is an isolate. The dowels also. I would purchase a commercial spawn if that is the worry. They are proven producers .
Strain isolation relates to sexual reproduction or spores. You would have to isolate a strain from the germinated spores then fruit it and see if you come up with a good fruiting mushroom that has all the attributes you are looking for.
Buy a commercial strain and match your conditions to that strain and your in business. It also eliminates time to grow grain spawn (2 weeks)

Cut a slice of dowel with myc on it using a sharp knife, transfer to agar. Once you are on agar you are running myc on starch energy.
Transfer the agar to grains, no added coffee.

No need to cut a dowel . Just drop on agar. Less contamination risk.

I got a LC KO syringe off of Amazon a few years ago and it was completely bunk I actually wasted a ton of time and money trying to get it to fruit. If your looking into starting a small farm it may be worth it to look into grain spawn from Aloha, I grow the K01 from them and it is excellent.
