Aa+ spores to agar

So last week I made 8 pasty plates and nocced them up with an aa+ ms. Only 1 showed signs of possible germination but it for sure has bacteria. Ima wait and see where. It grows a bit if there's anything to save but the other 7 had nothing. My question is can I noc up a wbs jar and take a sample from that and put it to agar to get some viable plates? Or should I just try to go ms to agar again?

in your opinion should I just do a tiny amount of grain so it colonized faster and I can make a few plates or should I make one jar transfer a little to plates then do g2g to the rest

You could
But that's defeating the whole point of agar.
You wanna make transfers from your original plate trying to grab clean mycelium each transfer until you have nothing but clean culture.

If you put colonized grain to agar you will still likely have bacteria to clean anyway.

I know I'll have to clean it I'm not worried about that my issue is I made 8 plates and so far have no growth. I don't wanna waste my time again if I can just drop some grain knowing it will have so e healthy growth

i only do grain-agar if im out if i dont have spores handy. it also shaves a
few days of the plates growth cause its already myc opposed to germinating spores. either way it gets cleaned before going to another plate.

Spore solution to agar tends to be messy and if you're not careful you can get germination (and possible bacteria) all over the plate unless you use an inoc loop to streak spore solution on the plate.

On another note, most newbies squirt an awful lot of spore solution into the plates instead of the standard one drop and this increases dramatically your chances of getting nastiness.

Spore solution to a small amount of grain and then cleaning grains on agar is actually a good idea, more so if your syringe happens to be dirty.

Since it sounds like you didn't transfer before inoculation your options are limited.  If you plan on putting the grain to agar you need to decide if you're gonna pick a grain as soon as it colonized and put the rest of the jar at risk, or wait for the whole jar to finish and risk obtaining a less-vigorous culture.

Also, I have had several dishes that didnt germinate and I simply re-used them problem free.  I just label them as blanks

I was thinking of just using one of my quart jars but only filling it like a 10th of the way just to get enough growth to make a few plates to clean up. I just wasn't having luck with that Ms to agar. 8 plates and only 1 might have healthy growth to transfer
