How is the mycelium?
    #23880671 - 11/30/16 04:52 AM (7 years, 1 month ago)

Hey
I've been working with a strain of Psilocybe cubensis, after a few transfers i had nice rizomorphic mycelium nothing amazing but it looked to be healthy and thick, i took sectors from these rizomorphic spots but now the mycelium has taken on a cottony appearance, and it looks very uniform.. is the mycelium senscing because of too many transfers?
I know very little about growing, feel free to correct me anywhere!
First picture is the healthy mycelium, the others are the product of transfers from the first.

The pictures aren't that good due to condensation but what I can see, your myc is fine for inoculating. If you want to be sure, make one or two further transfers but IMO you are fine to go for inoculation.

You have a nice evenly growth, don't overthink that thing with rhizo or tomentose growth. It's more about nutritions and other circumstances on your agar plate. A tomentose myc can grow rhizo once it's used for inoculating a jar.

Have fun!

Yeah sorry about the picture quality! is there a way to not get condensation on the plates?
Thanks for the replies, its cool it isn't to do with the amount of transfers but rather the media.. really interesting stuff! will be using these for grains now

To prevent condensation you should minimize temperature fluctuation as much as possible. You should store them at the temperature the room had when pouring them.

If you have to change the temperature (e.g. bc of putting them into the refrigerator) you should put them in an insulated lunch bag or styrofoam box or something.

Regards

In addition to storing them in an insulated container, allowing your agar to solidify after pouring with the petris stacked in a column will help prevent condensation.

Yes, what he said. Pouring in stacks of 10 is fine, gives you 8 good plates and two with a bit of condensation. You can get rid of this condensation in these two plates by putting a metal cup with hot (not boiling) water on them.

Yeah, pour from bottom to top plate. Watch YouTube how to do it

RR's Let's Grow Mushrooms series in particular is incredible if you haven't already seen it. He details pouring a stack of agar plates better than probably anyone else I've seen.

Weird that he doesn't mention it in the videos. I've seen him say it on here tons of times to stack them and put a warm coffee mug on top. Personally I just stack them, and if there's condensation leftover  a little extra water hasn't ever stopped me before.

OP your plates look alright to me. They fuzz up when there's different nutrient ratios in your agar. If ya took away some of the nutrients, i bet it'll rizmorph again.

Yeah, I also don't care anymore about the condensation on the top two plates. I have recognized that it disappears after a while anyway when stored in a Ziploc Bag.

Regards.

The stacking after pouring is the same concept the hot water thing is more for after the fact....op your cultures are probably getting lazy or you're not taking the leading edge.
