pasty agar tek preference

So I know Pasty agar is generally for people using SAB's but what about if you have a flowhood? It seems like if its done correctly it leaves less to chance than pouring petris in front of a hood even if the air is 99.99% sterile.  And I like the idea of a tiger drop instead of having to fish the wedge out with a scalpel to the grain jar.

I never did pastys agar tek, but I can't imagine why it shouldn't be able to use it with a FH.

Furthermore you can do tiger drop (Version 1 or Tic Tac to) with a petri dish too. Why not, just cut the tic tac to pattern in your agar petri and drop it into the jar.

I don't think I would be able to squeeze the petri to drop the agar. The tiger drop method can only be done with a container that is a) flexible and b) smaller in diameter than the recieving jars mouth. It's one of the reasons that I like the glad containers so much, few others have those attributes.

Yeah, I did it some times a bit modified, without squeezing, when I want a whole plate to be dropped in the jar.

I cut a circle around the myc part and lift it a bit with the scalpel that it's losen from the petri dish. Then I drop it into the jar.

Sorry for the misunderstanding, it's not totally the same like your tiger drop but somekind of

I could definitely imagine doing the drop if you loosened it up with a scalpel first, with a flowhood you'd definately have to drop it from behind the jar quickly so you don't blow in any contams that might be on the petri.

That's true. I already did this with bags sometimes. Cut a circle and a cross on the petri to have 4 quarters. Place it behind the bag/jar use the scalpel to drop.

I usually use jars and lift about a quarter of a dish with the scalpel to inoculate.
