Received my first culture, how to keep it going?

Hello everyone,

I received my first petri dishes in the mail today and set up my flow hood. I'm going to hit the lab work tomorrow. I've got a few questions on how my 'work-flow' should be to keep my cultures alive, this is what I've concluded from research so far:

1. Move some of my petri dish culture to test tubes for long term storage, let's say 5 to be safe. I'll revisit them down the road.

2. Let's say I have about 2/3 of the dish left. I transfer 1/3 to some to a new agar dish.

3. I transfer the last 1/3 of the dish to a grain master. From there I G2G into let's say, 10 other jars (to avoid losing viability). But then I use the jars and my spawn is done, Now what? :

In this situation do I take a wedge from the petri dish from step 2 and make a new grain master, and then keep that petri in the fridge and keep making grain masters as needed, then once that petri is almost done make a new petri just like it?

I hope to grow commercially one day and also wonder how the 'work-flow' of those guys would be.

1. ok

2. you don't need 1/3 plate for transfers. Just a small small amount will do for each plate you transfer too.

3. pretty much. you can make A LOT of jars via G2G...then use the cultures from your test tube to start another agar dish and go from there. I wouldn't keep the petri in the fridge because i doubt it would last since you can G2G for so long. Commercial growers use clean LC mainly though I do believe.

Went I have clean culture in my possession that I know has good results I'll transfer tissue samples to 8-10 new petri dishes. You should have 3/4 of your original "Master" culture left, leave that be, store it in the fridge. Once the other 8-10 plates grow out put the majority of those in the fridge as well as use one to make 4 Grains jars dedicated to G2G jars or bags. With those 4 jars you can make 100 jars or 25 XL bags. After your 8-10 plates run out you can make some more from the "Master" culture you still have in the fridge. This might not be the best method but it's how I've been doing things for awhile now, hope that helps..

Yeah G2G can make a crazy amount of jars. If you have 4 like liloldme said I would spread those into 40 jars - 10 receiving jars for 1 master jar.

Then you can take those jars, and if you wanted, use them all as masters and make 400 more jars etc. All within a few weeks you can have more spawn than you know what to do with.

So you use a 1/4 of your master plate to inoc the 8-10 plates?

Yes, even less if you're careful.

Awesome, liloldme and wowimflabbergasted, thanks for the help. I'm leaning towards using spawn bags because I've got both jars and bags on hand.. How much of a dish is sufficient to knock up your typical grain bag?

I would do 2-4 jars from one 100mm petri dish then G2G those jars to bags. Otherwise you really only get 1 bag per dish and that's no fun unless you're rocking tons of dishes.
