What agar sectors should be isolated
    #23875731 - 11/28/16 05:04 PM (8 years, 1 month ago)

Hi!

I'm a new grower, and I'm looking for some guidance on what I should isolate.

These Petri dishes have been isolated once so far from MS syringe. They were isolated from a single Petri dish.

The center might look a little strange cause there is an extra chunk of agar from the previous dishes.

Also, how do you guys think the isolations are going so far? How many more do you think will be needed?

Thanks!

they haven't been isolated. you've done transfers. if you want an isolate 10-20 more or so. I wouldn't bother with an isolate especially at your experience with agar level.

good work

Ahh i see. Thanks for the advice.
Well, I already have made quite a few Petris, have all the tools, and equipment. So, with practice I feel I'll get better quickest with continuing my agar work. Plus, it's a fun hobby! Ha

So, where should i make the transfer from?

Thanks!

So, Is a good rule of thumb to measure the distance from the center to the outside edge and pick the location with the farthest distance?

Does visual content factor into this at all, after several transfers have already been done?

Preciate the help guys!

EDIT: To me, 8-10 o clock on Dish #3 looks the best.

Haha I gotcha. I mean, like, does thickness, color, etc. factor in or should I just go by leading edge?

These convos really help me so thanks again ✊

Agar is kind of trickey to see and know.  In this pic I am not sure if I selected clean rizo growth or if the myc. went rizo to escape some form of contam.

That is why I love me a fresh fruitbody inner stem flesh clone.

Damn I don't have that eye yet lol. I just read some RR posts on isolation tho, and that answered a lot of questions.

I'm curious to see what other members say about your dish.
