I'm now wondering if my SAB procedure is fine. I just spray the inside wraps with Lysol, and put it on a towel. Cover the holes with another towel. Then after 5 minutes, I put on latex gloves. Wipe with ISO, wipe my arms with ISO, and get to work. I ISO the outside of everything before feeding it into the SAB through the arm holes.

Actually it can't be my procedure because none of my unused agar plates ever contam... could be my scalpel not reaching temps? Or maybe i took too long with the transfer...

Alright thanks do you prefer the squirt bottle Lysol or the spray? Also is the towel good or should I be doing work on the lid?

The towel should be fine as long as it is clean I'd use a white towel so that you can wash it in bleach before hand. I dont use lysol on the walls I use a high concentration of antibacterial soap and cold water( I find hot water fogs with humidity making it even harder to see). Once I have everything wiped down and in the SAB I then spray lysol in through the arm hole to kill anything I can that's left floating in the air. Then I let sit for 30 min or more to allow anything I can't kill in the air to settle and stick to the walls.

What do you cover your arm holes with? I was thinking of going with that magnet idea…

Actually you don't need to cover your armholes at all. Some guys here let unwrapped petri dishes with agar in their SAB until the inoculate them with open arm holes.
As long as you don't stir up the air infront of your SAB too much, there's nor risk of conatmination.
And don't wipe down anything outside of the SAB and then bring it into it. This doesn't make sense. Either put everything on the towel before putting the SAB on it or put it through the arm holes. And THEN wipe it down with alcohol.

Lysol is a surface sanitizer, not an air sanitizer. You need oust or something equivalent for anything airborne.
EDIT Apparently Lysol does have an air sanitizer, sorry, I wasn't aware of this. It has to be this type though...... Lysol® Neutra Air® Sanitizing Spray

I wait 30 seconds the water vapor will knock most of the spores to the bottom very quickly.
Do you use the same flaming techniques for agar transfers? Do you get no contams with agar transfers? If you don't, I doubt it's the scalpel thing.
If these are metal lids, do you still have the rubber seal attached? I'd recommend removing that..

Yes I use the same flame sterilization. Come to think of it, a good percent of my transfers get contaminated. I thought it had to do with unclean wedges. I use a pretty thick exacto knife, and isopropanol as the fuel for my make-shift lamp. The knife never gets red hot, only soots like heck. When it gets so hot it starts burning my hands from conduction, thats when I stop. So I dont know…

And yes, those are regular mason jar lids. Why would the rubber be removed? I've never heard of that

UHG I literally just bought a brand new bottle of lysol. Like, 5 minutes ago. Didn't see that tho. And last time I was there I looked for Oust too, can't find it anywhere.

I like to do both. Once before putting it in, and once right before using it. Thanks for the armhole tip, I'll keep that in mind.

My improve your sterile techniques explains it . And yeah get your scalpel blade red, as seen in that tek as well haha. Actually I'd recommend just reading the entire thing tbh. It will help cut down on agar contams. Also think about PCing your tools.

Yeah. I thought it was suspicious at first because it was a lot 'grayer' than the mycelium I sew elsewhere. A couple days later trich developed very near it. (or if it was already trich, it just turned green and sporulated).

Do yourself a favour and get a butane / propange torch. At least these 5$ creme brulee torches.
The soot is not your problem, your problem is that the blade isn't getting sterile in your lamp fire.
These torches are great, most of them has a locking mode so they are firing the whole time you are doing SAB work.
You won't have any more problems
