Reasons why spores don't germinate / no colonization

Hi

I inoculated a set of 4 BRF jars with a GT spore syringe. 9-10 days later I saw growth in all the jars. A week later I inoculated 5 BRF jars with a B+ spore syringe (same vendor) using the same technique with a couple of differences.

1) I didn't use micropore tape over the inoculation holes.
2) During the sterilization in the PC, I think the water boiled up more because the lids had alot of moisture between the lid and the foil; even some droplets. I had some verm also stick to the needle when i inoculated which I hadn't had happen in the first set.

Visual inspection suggest the moisture of the jars to be normal from what's visible, no puddling at the bottom or anything.

It's been over 12 days now, and there isn't a single sign of growth of any type in any jar. No mycelium or contaminates visible.

When I had tried my first grow last year, the same thing happen (B+ as well from another vendor though). I had zero growth in the jars.

Is water getting into the jar through the inoculation holes a possible reason for 0 growth in a jar? what are some of the other causes other than a potentially bunk syringe)?

older spores can take up to 3 weeks, 12 days is less than 3 weeks, thats why.

Give it time. Spores from ms syringes can take up to three weeks to germinate.  Its a crap shoot with ms syringes. I say learn agar so you can clean up cultures and isolate good growth. Check out pastywhite' easy agar tek

ok..I hadn't heard the 3 weeks before. I've got some agar jars ready but had forgotten to throw a drop of the B+ in one.

I'm just going to leave them anyway, but was worried a bit about the condensation under the foil.

ya, followed the tek. Foil came off prior to inoculation and stayed off.

What about your tape situation? After inoculation, you didn't retape the holes, did you? (Did that on my first PF Tek and couldn't figure out why colonization was taking so damn long.)

mp tape is breathable, you shouldnt be able to spot a difference in colonization time,
I suspect something else was going on, changed temps, bacterial infection etc.
especially bacteria can make jars take damn long to finish. looks all white, just has a slightly different texture.

But, if masking tape was used instead, that can definitely cause problems.

On this batch, I didn't use any tape at all. I'm hopeful it's just really slow germination at this point.

Did you get um to wet? gt's work great, that b might be contamatory giving it a hard time?  Try some pour agar and hang up the pf.
