Ok soo i think i may have bitten off more than i can chew...i recently traded for a lamanarflow hood and alot of edible gormet wedges about 18 to be exact and was wondering how to go about storeing these i would eventuallly like to fruit and collect spore prints /untill i can manage that monumental task how would i keep the myc vaible? My plan was to make at least 2 dishes from each wedge(do i really have to change agar nutrients every 30 days ?) Should i store them in fridge after colonization?would a slant last longer?

If agar is poured thick and dish is sealed with parafilm they will keep months in the fridge. Slants will keep longer, I use 50 ml  tubes with 25 ml of agar for slants. The man problem is for the culture to dry out.Some are storing wedges on sterile water for extended time with no refrigeration.

I use 1/4 pint jars with thick agar and toothpicks, works well. I also store backup cultures in sterile water.

According to GGMM the wood seems to provide longevity where agar alone failed.  I just finished resuscitation and passage of my entire collection that was put on antibiotic MEA slants with toothpicks in October 2014 and every one survived. Prior to putting them in the refrigerator I opened the caps 1/4 turn and wraped with 3 windings of parafilm and put them in a ziplock bag. To resuscitate I just pulled out the picks and dropped them on MEA.

P.S I bend the picks in half before wedging them in the tubes then rotate the tubes while cooling so the don't get entirely submerged.

Thats the exact same findings I have achieved. I am temped to see what pink does on wood at lower temps.

Has anyone stored cultures in LC.?

Was wondering this since venders will send syringes out and mine have lasted over a year at room temp

lc is a nutrient souloution which tends to go bad after a while ime should i store emmediatly after making in refridgerator?

You can store cubes on agar slants. Only wood lovers will benefit from wood in the slant.LC aren't ideal storage. If you go with liquid just use sterile water and store wedges in water filled tube.

Cubes will transfer to wood and from wood, not ideal. However if you watch Gr0wer's video you will see he uses them for other reasons also.
One of them which I think is a great idea is to hold down the agar in place. Removal is also easier, limited agar contact.

I was thinking to guarantee clean storage in water you could do no pour agar in 1/2 pint with an LC lid.. Add a wedge to let the dish colonize then inject x ml sterile water through the LC lid, shake it like crazy then draw the water back up and store in sterile centrifuge tube, test tubes or whatever else you want to use..

So is this how most people store their cultures?

I recently got a bunch of cultures that I want to make sure I don't lose.. My plan is to get the LC syringes on agar.. After its clean I was going to drop a bunch of small wedges into waster like the tek suggests as well as make thick agar jars like Gr0wer for the fridge..

Is it safe to leave the cultures in water at room temp for years? Or is it smart to take each culture to agar ever 6 months to a year then back to storage?

From what ive read the sterile water puts the culture in suspended animation of sorts. Especiallly with a layer of mineral oil on top of the water. So no refrigeration is needed for years. I ran into a few of my water cultures to contaminate so now i use them as a last resort backup and run the small half pint jars with about an inch of agar and toothpicks as my go to masters. Also, always have multiple masters especially if your a production grower like me. I have 3 jars of each and 3 water cultures of each of my main strains.

The oil will also help prevent oxygen dissolving into the water.
No 02, no nutrients or energy = stall, dormancy.
