Contam free agar plates?

Pulled these B+ cultures out of the fridge about 6 hours before taking these pictures. Was thinking about making wedges with the plates and inoculating some jars of whole oats in a day or two for an experiment. To my untrained eyes, they appear to be contaminate free? I assume tomentose myc is ok for inoculation? Thank you in advance for your thoughts and opinions.

They look good man. I think you have tomentose growth because you just have a shitton of strains growing together on those plates, further transfers would help with that. But anyway, tomentose growth means nothing, it can become rhizo if conditions change.

Rhizo growth comes in with low nutes those look fucking really good i would make some agar up with lower nutes in it to get some rhizo so you can see all the active strains ... id take 3 small tramsfers and drop the rest into grain

Got a case of 500 petris coming any day now. Didn't think to make transfers from those before inoculating. My dumb ass was just gonna start from scratch. Good suggestion, I will do that. Will also tinker with my agar recipe. It's a premix, and am still trying to find the sweet spot, ratio wise. This time I'll try 40g/L instead of the recommended 50g/L. Thanks!

With a premix the problem is, that the Agar will lose thinkness when you decrease the conecentration of the premix. If you are happy with your agar thinkness at the moment, better don't change the concentration. If I were you, I wouldn't change in thinknesss for the sake of tomentose/rhizo growth, cause it's not that important (as I also thought at the beginning).
Next time you better go for buying every ingredient of the agar you favour and mix it yourself!

One advantage of premixed agar is that the plates are clear after pouring without sediment or something (premixed MEA). If you don't care about sediment or cloudy plates, mixing your agar is cheaper + you can adjust nutrients and agar content.

Making my own mixes is something I would like to explore in the future. There are definitely some advantages to doing so. For the moment, I have a pound of premix, enriched MEA to burn through. It's a great product, and has served me well so far. Seems to always set up super stiff at 45 - 50g/L. I have read that spore germination is faster on a looser growing medium. Not sure if it really matters for transfers though. I hope to have all the finer details of agar work figured out by the time I burn through a case of plates. Experiment time!

It does not matter for transfers. A higher nutrition content slows the growth a bit. Can you post the detailed ingredients of your premix?

Not sure where you live man but you can buy agar sticks at an asian mart for like 1.30$ for 20 grams ill buy 400 grams at a time then potatoe flakes or malt for cheap the price youre paying for premixed is enough to buy 6 months worth or more of doing it yourself... that dude is right about tomentose and rhizo its not important at all but with rhizo its easier to see sectors and that i do 20 grams agar 10 grams malt with 1000ml and 10 10 to 1000ml for spores

I have a couple asian markets in my area. I didn't know agar agar came in 20g sticks. That's awesome! Always just seen packaged flakes for way too much money at the grocery. There are couple of beer brewing places I could check out for malt. I assume you just drop the stick in water to dissolve? I'll log that recipe for later use. Thanks.

I blend them in a ninja you can do it in a coffe grinder or just drop them in it takes longer tho and they come in 2 10 gram sticks in a pack in my asian mart the sticks are in a different aisle then the powdered shit but ya my chick bought some for me at whole foodd it was 13 bucks for like 7 grams or someshit ... also those sticks are precolored
