How much Agar culture needed to inoculate 1 x quart jar of WBS?

After coming to the realization that if I'm to become proficient at mycology then I will have to move onto agar instead of ms syringe to grain I'm off to buy supplies to make some agar.

I am going to follow Pastyplates tek using glade mini rounds. I am wondering however once I can see a healthy mycelium culture how much I let it grow out before cutting out to inoculate a quart jar of WBS.

Do I wait for the whole container to be colonized or can I just wait until it's say 1/3 grown into the agar and then cut out to inoculate a quart jar?

Or wait for whole thing to be colonized and then perhaps split between multiple quart jars? Which raises the question how many quart jars could a fully colonized glad mini round knock up?

Also I'm assuming once inoculated I would just store the agar container at room temperature with ambient light?

You can cut any size wedge you want out and put it in the jar

You only need a little wedge as big as a grain of rice or two.  If you use more, the jar will colonize faster.  You don't have to wait for it to colonize the whole plate, you can make a transfer as soon as you're sure the mycelium is healthy and you have enough to inoculate all jars you're planning to inoculate.

Awesome thanks for the info. Also just wondering for ones that show signs of contamination could I just pressure cook them and then dump out the remnants and reuse or just toss?

What "ones" are you referring to? Agar plates or grain jars?

If an agar plate has contamination, you can transfer a tiny bit of the healthy mycelium to a new agar plate if you want to keep that culture going. You will generally have to do this anyway to obtain a clean culture, since your source is probably not totally contaminant-free. This is one of the main goals of using agar, to let your culture AND its contaminants grow so that you can use transfers to selectively extract and grow out just the mushroom culture in a clean setting.

If a grain jar is contaminated you will just dump the contents into the trash and wash the jar for next time.

For the most part, you never use or re-use anything that is contaminated. In general you will throw it out, keeping the contaminated contents outside and/or away from your growing and "lab" area. One exception being that you can fruit or spawn to bulk cakes or grain that have a little bacteria. You don't need to pressure cook before dumping out contaminated stuff, but you can if you want. I wouldn't bother sterilizing the contents if it's just green mold or some bacteria. If the contamination is really nasty (purple, black, rainbow, etc) then definitely be careful to not breathe it in when you dump it.

Ok thanks. Yeah by ones I meant the agar containers which in this case would be Glad minis.

Also to confirm while germinating colonizing do I just store the agar containers at room temperature with ambient light?

Room temperature is fine.  I don't think light has any effect on them.
