Quick agar question

Hey folks, I'm going to start cleaning up cultures today on agar. I have MS and clones. About 27 plates. My question is: How many samples should I take from the donor plates when doing transfers? It seems like people take one from each plate, transfer to the new plate and then discard the donor. Is that correct?

there are a lot of ways to answer this, and mostly it comes down to what ur plates are looking like.
if i have 3 MS plates, and one looks particularly healthy, I will only transfer from that one.
its a roll of the dice.
keep ur labeling accurate in any case. very important, especially down the line.

i'd probably take no more than 3xfers.
depends what else is going on….do i have more projects going that get plate priority, am i more interested in exploring MS grows vs.clone grows, etc.

It depends what your goals are.

I would transfer the fastest / best looking sectors to new plates and see how the selected sectors perform.
You need a lot of plates if you take more samples after each transfer.

Before tossing a plate transferred from, I'd wait until the new plate is grown out a bit, so that you can see, that the Transfer worked well and there is no contamination on the new plate. When you have a clean new plate, toss the old one (if not needed) and go on tranferring from the new plate.
In most cases I only make one transfer from a plate, don't forget, that every opening of a plate is a contamination risk. So if you have a very good plate and don't plan to toss it anyway, just make one tranfer and use the old plate for grain inoculation or whatever. To many transfers increase the risk of loosing the plate.
