G2G question

I have several quarts of WBS that is fully colonized but want to switch to using rye berries. Is it okay to put the colonized WBS in with rye berries. I figure it should work just looking for confirmation before I do it.

It depends, how did you inoculate the wbs? If with agar then you should be good to go.

What difference does this make? I'm still (always actually) learning.

Well you wouldn't recommend someone to go bulk with spore syringe as the inoculate right?

I take it this is 100% colonized, it's solid all the way around for about a week and a half

Why you say that? I have large holes filled with polyfil in them. I just put the coffee filters on top of them because I have several cases of them stacked on top of each other, and I write myself notes on them. But they should be 100% colonized now?

No need for a consolidation period with grains. When they're fully colonized they're ready to spawn to bulk. Yes you can mix different kinds of spawn jars. You could even go as far as mixing your rye and wbs in the same jar with popcorn before PC. Myc won't discriminate.

yes, I guess your WBS is already a mixture of grains anyway. I have noticed it can take longer for different grains to take off when mixed, I can't recall the term but its that they are not used to the new grains and so have a slight delay getting used to them. I have noticed this also with the exact same grains but at different moisture contents, like adding pretty wet colonised popcorn grain to drier grains of the same popcorn.
To test if the grains are clean I will sometimes shake them up to see if they recolonize in a few days, it is no guarantee though. If they do not colonise again they are most likely contaminated and so would have fucked up all the jars you transferred to.

Alright now this shit is getting more complicated than I thought. So I should shake them up and see if they recolonize before I start. Last thing I want to do is put in a lot of work and lose it to contamination. I am also looking at agar and Petri's on eBay to buy at this very moment, for sake of not looking it up what is a good type to start with? I am seeing soy agar, potato agar, seaweed agar, and I figure it's best to go with glass petri's.

My favorite is malt extract agar (MEA). Working with glass petris is a PITA.

I like both mea and plastic petris. I've only used them so I can't really compare but I can think of some advantages. I go through a lot and it's nice to not have to add an extra step (sterilizing dishes) when I'm already busy enough as it is. I could just throw them in with the agar but it's still an extra step I don't want. Plastic petris I just wipe down the packaging with iso and cut the bottom and slide the plastic off. Plastic dishes are also clearer than glass, believe it or not. Glass is slippery too. Plastic is cheap and can be recycled. Mea is a good well-rounded nutrient agar to start with. I find it suitable for all my needs since I'm not doing any long-term transfers right now, but if you want to do that pick another mix and switch it up to keep the myc vigorous.
Whatever you choose, have fun with it. You probably won't be able to stop yourself anyway lol

It is a 'pain in the ass' You have to clean them every time. You have to sterilize them...
Sterilized polystyrene petri dishes are comfortable to work with. You can not use them multiple times but it is easier to work with them.
