Agar Plates from Spores not Growing?

Recently I purchased several different spore prints of cubensis, and I had a syringe of Panellus Stipticus culture. I poured some agar. I innoced three plates with the Pan Stip syringe. Then I proceeded to innoc several (like 6 or sometihng) plates with a innoc loop I made from some 28 GA wire I have. I flame steralized it with a BIC, then cooled it on the agar. I scraped some spores onto the loop from each plate then put it on the agar. Enough to see a rather large collection of spores on each plate. The prints were from different vendors. Neither grew, except the Pan Stip did. In fact, all the pan stip plates got great growth with no contams. The spore plates, some got Cobweb mold and none actually grew. Wtf.

How old were the prints?
I've had it happen, finally decided the print was bad. Old in fact I found out later to say the least. Also were taken on paper not ideal

One of them was from 10-11-16. I can't remember the other. They were both pretty reputable traders.

not sure why those sporeprints aren't working out but maybe you could try using sterilized cotton swabs instead of the loop. and/or maybe some type of streaking techniques. and could it be possible that the panellus stipticus was an LC/LI that was made from a clean culture? i notice alot of non active mushroom species are sold that way as opposed to just straight spores.

Yes it was an LC. Quote:

I have BOTH PDA and MEA plates from Seaweed Solutions on Amazon. Mixed with the amount of water labelled on the thing. I think it was 1.8L water/100g of MEA and 2.5L water/100g of PDA.
As for the loop, how could I use it properly? Should I buy a proper innoc loop off amazons instead of making my own?

I've had spores take close to a month, give them more time before giving up on them.

Prints are not guaranteed to be contaminate free because they are not usually prepared under sterile conditions. It is possible, but unlikely that both of your prints are nonviable. Scooby, I smell a mystery coming on.

But both? I got them from two separate vendors!

I've had spores take two weeks to germinate on agar before. 7 days isn't worth getting worried over. Be patient -you'll know soon enough if your time was wasted or not.
For now- be cool.

If you can't get it going on agar try injecting a small amount of grain with spores as it will be able to absorb much more solution than agar. I've had to do that a few times. Once it get's going grab some of the colonized grain and put it on agar to clean things up.

I'm in Southern California, so the temperatures have been ranging from 14°C (57.2°F) at night to like 30°C (86°F) during the day. But I keep them inside, on my desk, so the temps are somewhere in between. I keep them in a ziploc bag
