Cloning an invitro pin VS isolation on agar - which is best?

I see a couple different approaches with isolates... some seem to take an easier route and let pins develop on their petri dishes, and then clone the pins, other do strain selections via sectoring and then fruiting out the isolates.
What are the advantages and disadvantages to each method?
Invitro takes less time and expense, and give you a clone with an obvious thirst for life... Plus I don't have to make a bunch of minicasings to fruit out 12 different strains.
2nd Q- do I need to fruit the isolates out int the same substrate they'd eventually be in- like coir and verm, or can I just fruit the isolates out inside a jar on a brown rice cake?

You answered your first question. 2. Isolates can perform different on different substrates. If you go with the isolate route with that much effort I would recommend to use the same substrate that you want to use later. (coir/verm)
An easy way is to clone a nice fruit, eat and test for potency and if you are happy with the result use the clone for future projects.

Isolates look pretty but can be the lone gene strain that doesnt fruit and so it will just sit there doing nothing.
When you clone you take a multi group of genetics but they will fruit and you should get mushrooms similar to the one you picked. To isolate down from there is lots less transfers than from multi spore.
