Hello there,

So yeah, I was out of here for almost half of my life and for some reason I was missing the hobby so I just gave it a shot.

This is my first agar project (back then it was for professionals only). Here's the setup :

13-15 years old prints that were stored at room temperature, dry, dark for all that time. Used 2 different prints.

6 plates were inoculated.
2 BRF jars inoculated after making a syringe and wait 2 days before
poking holes.

Results so far :

No growth at all after 15 days.
No contams.

In my book : the prints are dead.

What are the odds of being proved wrong?

Maybe you can try some forced hydration, that's pretty old though I wouldn't know the chances. What kind of genetics you trying to revive, anything interesting?

I would just go ahead and get a 10$ syringe and try the old stuff in the meanwhile

They probably are dead. Even the great agar master RR used a whole print and had to mix it will others cubes spores just to live. Look up how the rust spores strain was revived it was only 12 years ago in that story. I agree just spend the money on new syringes. One spore syringe form a sponser can make a lot of germination plates. After first grow you can clone your way for a year.

It depends what 'a long time' is for you. I had success with a ~ 8 year old spore print. Old prints need to be hydrated a lot to germinate. IME unhydrated spores -> agar doesn't work if the print is too old.

Let the syringe sit for another week and try it again. But 15 years... I don't know. Good luck :syringe::thumbup:

Damn thats awesome. So put old spores into syringe, let rehyrdate for a couple weeks? Then try on Agar? amazing....

I would test the syringe on at least 10 agar dishes. 5 with a low agar content to provide extra moisture for the germination process. It all takes some time. If you really want to get it to germination I would do it as a side project. Meanwhile you can use a new syringe.

Thanks for sharing opinions.

I scraped two different strains (you know, the microscopy strains) on 2 agar plates (4 total).  Once the syringe was ready, two days later, I put a couple ML on some other plates.

The main problem here is for long term storage was room temperature during the whole time.  Even then, I believe 15 years old is mostly a waste of time or a side project / experiment only.

I still appreciate my project being my first try at agar and first try working with a flowhood and my sterile techniques look adequate so far.  15 years ago the kid that I knew was a little more sloppy.

Yeah my plan A was to get my hands on something fresh for a new start but then I recently discovered that living North of the US border is no longer easy to buy from a vendor.

Still looking for a good source...

You try the free spore ring. A few bucks for prints is great hard to get places like your doing.

Yeah I sent a sizeable donation to free spores ring Earth last week !  Will take a couple weeks though so I'm still looking for a vendor.  Think I found one.
