agar cultures in fridge to grain
    #23786729 - 10/30/16 08:04 PM (7 years, 2 months ago)

I read somewhere that its not optimal to put a culture that has been in the fridge straight to grain, you should always make another transfer to agar first just to make sure its still clean.  Is this feasible or can you put it straight to grain if your sterile technique is on point?

It's a good idea to make a transfer but it's not 100% nesicerry. I just finished knocking up 8 bottle subs with a plate I took out of the fridge this morning. I've done it lots of times and I can't think of a time it caused an isue.

I mean... it depends whether the culture is clean or not. The challenge is that you don't really know, but the longer it's spent in the fridge, the less likely. Depends on other things too; if you have a dedicated mycology fridge that's super clean and doesn't get opened much, well that's different than your kitchen fridge. General rules exist for a reason, but nobody can predict the state of a specific culture that's in your fridge right now. Give it a try and you'll learn something about the environment you're working in.
