Pleurotus ostreatus (Oyster) Grow --> Agarplate contaminated or ok?
#23776285 - 10/27/16 10:51 AM (7 years, 3 months ago) |

hey guys,
besides my cubensis grow experiences I want to grow some oysters
I bought from a laboratory in my city a liquid mycelium culture in a syringe and put some LC on agar to multiply it - don't want to buy a syringe everytime its empty.
The LC in the syringe is definetely sterile, cause it made under laboratory conditions. The needle was also sterilized before first use.
I put some of the LC on an agar plate (LME-Agar) but unfortunately I dropped more LC than I wanted on the plate.
The result is the following:

As you can see the mycelium grew on every spot on the agar. Can you see any contamninations? The mycelium is quite fluffy, but I think this is due to the fact that the humidty inside the agar plate is quite high because of the "to much dropped" LC.
Is it still possible to transfer from this agar plate to other agar plates for multiplying? If yes, which part would you recommend to tranfer from?
Or shall I use it directly for grain inoculation, because tranferring is not necessary/not possible anymore?
Would appreciate any help

Looks ok? I would take and transfer to another plate. The first pic lower left about 7-8 o'clock !looks good for a transfer. Then you can really tell. just take the leading edge.

It's good in this case, it means it has strong growing genetics. Far too thick and radial to be bacterial avoidance.

Ah, ok, fine, sorry for my stupid understanding
So the triangle I drawed on the picture is good for transfer?
Or do you think I should do both:
Transfer the triangle to another agar plate and also take a wedge and directly inoculate some grain jars?

Have you checked for bacteria pools or unusual colours in the agar? If you have, and to me it looks healthy, transfer away.
Just keep checking for anything odd.

Actually I also can't see anything looking strange on the plate and the free of mycelium agar spots also look well.
"Transfer away" on agar (for further cleaning) or also to spawn, what do you recommend?

In that case I would do both, the agar transfer will be backup. Good job, I hope you get some strong genetics.
Good luck, Ferather

Yes transfer the triangle. It looks like good plates ,but that triangle is more normal mycelium growth. It should continue looking like that after transfer. Keep the plates and wait and see if anything changes.

"more normal mycelium growth" = good growth?
So: 1. Transfer Triangle 2. See what happens on new plate 3. Let the first plate colonize further and drop wedges to rye spawn.
Correct?

That would be my plan those two plates are probably usable give them some time . The new transfer should be good for sure , time will tell.

It's the same plate but two pictures
But thank you, I let it grow another day until dropping it to grain I think. The plate hasn't be fully colobized for rye drop, right?

You can drop on grain anytime. If there should be some mold there you just waste some grain.

Just do both, one to grain, one to agar. Leave the original to grow. If it where dirty the longer you leave it the worse it gets.
However, this is scaremongering. It looks clean. He checked for usual signs of bacteria.

Yes, acutally I can't see any contamn, and I know how bacteria and mold look on an agar plate, since I hade these issues before(eventhough all in all my no-contamn rate is thank god quite good).
So tomorrow I'll do the transfer for multiplication and the agar wedge drop to grain. I think I will also make a second rye jar to inoculate directly from the mycelium syringe of the laboratory. Just to see, if it's also possible without contamn.
Maybe I loose some rye with it, but rye is the less expensive thing in this hobby so I think I can afford it :P
Thank you again for all your help.

I have colonized some rye jars with oyster mycelium with agar.
I'm not really sure about one jar.
Do you think its contaminated?
Pictrue 1 - Three days after inoculation with an agar wedge. Unfortunately I wasn't able to get the agar wedge sticking to the glas so I was forced to "roll" the jar a bit so that the agar wedge was covered with rye berries. This white stuff on the top could therefore be mycelium which got there because I had to "roll" the jar a bit, so the berries there were inoculated with mycelium. But it also could be cobweb.

Picture 2 - After Picture one I shook the jar. Picture 2 is 3 days after this shaking.

Picture 3 - After the picture I shook it directly.

What do you think guys? Doesn't look good, hm? (don't forget it's oyster mycelium, not Cubensis)

Hey mate,
thank you very much
I wasn't really sure, thought it could be mold or some bacteria. I still have to learn to see the difference between cubensis myc and oyster myc, if it were cubensis, I'd toss it directly

It's clear, so its just the mycelium producing heat, and the water cools on the glass. If it where yellow or any other colour, or even slime, you have issues.

Here a picture of my oyster grow in "bags" with straw pellets.
It's going really well, I think next week I can start fruiting them by cutting some cuts into the bags.

Thank you very much, I really like that hobby and because of that I try to do my best for good results
I wait until the mycelium forms the first bigger nodes and than I put these bags in a slightly cooler room. Then I'm going to cut some slots at the points of these nodes, so that the fresh air exchange forces the mycelium to build some fruits there
I hope I can start doing this next week so in about two weeks I'll eat my first oyster pasta

If you haven't already you should put some holes in the bags. They need air or they can go anaerobic. You can use a arrow head or knife to put holes in it.
