Hi everyone, It's my first post on sroomery I'm trying to make a liquid honey culture. My substrate is a rye grain so to make lc I used a soaking water from rye. 200ml soaking water 8ml honey
First I sterilized only the soaking water in pc (15psi) for half an hour then I mixed the soaking water with honey and sterilized for 20min more.
This is how look my jar.

Are those floating flakes fine? thanks

Those 'flakes' could be caramelized sugars or even proteins from the boil/sterilization. BUT, I've done a few experiments in my lab and there are also definitely organisms that flocculate into similar chunks. It's hard to tell from the pictures, and considering that honey is unusually good at keeping bacteria at bay. I'v never tried to make honey LC for mushrooms or anything, but I do work w/ microbes, so take anything I say w/ a grain.

I think it's not caramelized sugars or protein because I noticed those chunks after a few days and it's look like it's still growing up :P

Too much honey. Not good. Straight grain water lc does just fine, honey is the worst at making a jar clear and clean. How are you knocking up such a lc. If you start form spore syringe you might be out of luck. If your not starting out with agar work id start with that. In the archives there the poor mans agar tek for those who can't find agar easy. That will start you off until you can find agar and give you growth to cut out when do fine it.

I was following this tek https://www.shroomery.org/forums/showflat.php?Cat=&Board=2&Number=10446142&PHPSESSID=&fpart=1#10446142
Quote:
4 ml of honey per 100 ml of water (around 4 % honey solution).
so I though 8ml of honey will be fine

I used a spore print and I was doing everything in my glove box. I'll try one more time with destiled water.
About agar. How should I store agar after injection?

Spore print or syringe doesn't matter, unfortunately either has high chances of contamination if taken directly to LC.
Also you're going to want to use a SAB instead of a glovebox: https://www.shroomery.org/forums/showflat.php/Number/20048771#20048771

Now if you really want to use straight spores form a print you could always make it into a syringe and then divide with a high of sterile water like 500 ml of water. It reduces the chance of a contam spores with still a fair amount of cubes spores . It's a lot of work for 97 percent success rate. Less work is to agar plates and use a sterile swab to dab in the middle form the spore print. Cut away the clean grow into new agar plates.
