I think he means that you should pour the agar and let it cool while the jar/plate is tilted. The agar cools and stays in the tilted position. If you put your jars/plates on a straight shelf or something the water can pour to the corner.
The idea is not bad but the water pool is a breeding ground for bacteria. (bacteria is almost invisible in water for some time.)

So before it even gets pc'ed? Cause from what I have seen some bacteria looks just like mycelium growth.

If you want to use the 'tilted method' with Pasty Agar tek the containers have to be slightly tilted inside the PC until they cool. Not very practical. I doubt it is worth the effort.

Yes this is what I meant, it pools in the corner. Some people put jars into an SAB and pour the excess water off, but I still find some always remains, this lets it always collect in the corner. I often test sterilization techniques by storing jars for plates for months, some have been inadvertently kept for several years after I forgot about them.

If your plates are level in the PC, and you leave them to cool in the PC, then you could tilt the entire PC at the end when it is cooling down. The agar will be liquid for quite some time. Only a slight tilt is needed, you can prop the PC up on some crumpled up tinfoil. More often I microwave mine for just a few seconds until it is liquid in some spots. This can be enough to dilute the "free water" and have it mix into the liquid agar and then set again, so no tilting is needed. Or it can be microwaved (or put in hot water) again and then leave it cool while propped up at an angle

Bacteria looks like bacteria and doesn't look like mycelium at all... it just changes the way mycelium looks. If you're looking at pictures of bacterial substrates and seeing the white shit as the actual bacteria then you're misinterpreting why it looks the way it does.

I ran out of lysol and instead sprayed some air deodorizer in my SAB. I think that was a bad idea.

you dont need lysol or deordorizer or anything like that. just soap and water.

So plain tap water and liquid soap (50/50 solution?). Spray on the walls of the sab and on the bottom (i.e.: table counter, towel), then wait 4-5 minutes for everything to settle down?
The plates that are sitting in my closet haven't shown any signs of contamination yet (Thursday would be 4 days post-inoculation). But when I hold the plate up to the light and squint a little at a certain angle I see some dark coloring underneath germinating mycelium. Being super paranoid I'm thinking these plates are DOA. I've cooked the agar properly, I pressure cooked it properly....the only bottomneck maybe my sterile procedure in the SAB.

yes. maybe not a 50/50 mix, just enough to get it sudsy ya know. the idea behind the wet walls in a sab is that any contams floating in the air inside it get stuck to the walls instead of bouncing off of it. its very effective, and was first noted by the same scientist that created pasteurization.

Over time I find that the lid becomes easier to remove. Is this something I should be concerned about? Is it time to buy new pasty plates? (glad mini rounds)

The containers start to deform after time. That's why I don't like to use PP containers. You should buy new containers after some uses.

if it is just slightly loose it should still work and pasty says he prefers them that way because they are easier to work with.
there still will be a complete seal with the looseness unless badly deformed.

Assuming Long Term Storage?
And if so, then yes. I use pasty plates to store all of my cultures i want to save for later use. I just pour them a little thicker than normal, let the growth reach the edge, then put them in a ziploc in the fridge.
thanks for your answer.

Yup. I've used pasty plates after more than a year in the fridge. Pour them thick enough and they won't dry out for a verry long time.

The higher the temp/pressure then the more likely they are to deform, if you have screw on lids and have them tightly screwed on the will certainly deform. Depending on the ingredients you can get away with just steaming agar. If you do have agar requiring a PC, or are just worried then I advised before that it could be PC'd in a glass jar, poured into PP containers and then just steam them or PC at a much lower pressure.
I sometimes do this with grains. If you PC at 20psi+ it really ruins most PP containers, the thinner the wall the worse it is, and some shapes are more prone to deforming than others. I have also "repaired" containers by adding thin layers of silicone sealant in the grooves of the lid so they get a tight seal again.

I use single use petri dishes but tried to use PP containers for Grains. I stopped after I had some problems with deformation and contamination because of the deformed lids. I have one container next to me. Maybe I give it another try. Last try was a few years ago with a very thin PP container. This one is thicker:

I use containers like this as trays.

I have gotten ziplocs up to 25 psi without too much issue but I don't recommend it lol.
