agar growth, refrigerator and vigor.

Hello,
I noc up myco bags every 3 weeks or so, with a LI syringe.
I make the syringe with a blender and an agar petri of Golden Teacher.
My current procedure is to grow out a strain to about a dime size on a 90mm dish and put it in the refrigerator. When I take it out, it takes about 10 days for the new growth to reach the edge.
I feel like the growth is vigorous at that point, but I recently read that someone likes to grow it out with a few transfers before using it to noc up spawn.
I simply don't want to be doing agar work in my SAB every three weeks.
Thoughts? I'm watching a bag colonize well from my current procedure of cooling and room temp growout... but I'm still new to this all.

i do agar work a few times a week though i use a flowhood so it's much less of a PITA.
personally i think it defeats a LOT of the purpose using agar if you don't do a few transfers before putting it on to a substrate.... you're just growing everything in the syringe/print instead of being sure you've got 100% clean culture. might as well go straight spores -> grains if you're gonna do that. plus, transferring good looking growth gives you better chances of getting high performing strains. there are too many reasons to do transfers for you to not be doing them imo. even if you only do 2-3 its a lot better than doing 0.

My question was poorly written. These petris have many transfers, the one I'm using is on its 7th. Its just that I cool them to stagger out the noccing dates.
Does that make sense? Sorry for the confusion.

meaning I cool the last transfer, but I let it grow out to confirm contam free growth.

ohhh gotcha. yeah what you're doing should be completely fine... the growth should be no more or less vigorous if you do/don't do an additional transfer after removing from the fridge. and if they're colonizing the plate in 10 days, you have plenty of vigor you can affordably lose anyhow.

Awesome. Many thanks for hearing me out. I just looked again at the bag and it looks great. I've been chasing down a spawn contam, and I think it was my whole LI syringe prep/blender procedure that sullied my clean agar plate. Still ~ I'm fighting to have the mycelia as strong as possible, I just recently heard about the cooling taking more than a day or two to recover.
and I've been at this for 6 years!

i see. i'm sure it does take a little bit of time for the culture to rebound after removing from the fridge. thats the whole reason store slants there long-term, after all. should be good as new once it wakes back up, though.
the vector could very well be the LI syringe. i try to avoid syringes whenever possible, and only use them fresh out of the PC. another thing it could be tho, is the fridge its self. fridges are one of the dirtiest places in a home... i mean how often do you clean yours?

The petris all go in a ziplock that gets cleaned thoroughly before going in the SAB.
I think I'm chasing this contam down fast. I cannot wait to get more than one flush.
