Hey everyone have been just reading for awhile now and just decided to repost.
I've been very hesitant on posting. Not sure why. I think I've hit a roadblock where I just can't get anything clean. I constantly inporve my sterile tek in my sab. Still something just happens super discouraging.
Besides that hello:)

What has your technique been? We can only help with information. Pictures help too.

Sorry I want really specific I was more venting and just trying to get that first repost out there.
In my sab I use saltwater leave wet.then i "shower shit shave" come back into my room stillvwet towel wrapped around me. Medical gloves dust mask (just because) 70%iso.
Am using rye. Just started trying out millet for the mass inoculation points and basic agar. Everything PCd for 90+ mins I usually sit around after my 90 min timer goes off
I currently have a few plates I cloned from a pin that I cut and dropped a piece into my rye ( I couldn't not tell what was going on in the plate) and I have healthy myc.
I just recently upgraded to sfds to make my results more uh accurate less variables.
Sorry if this is a little jumbled I got like 3 hours of sleep last night after a 11 hour shift.
Thanks for the quick responses

I would change your technique to shit, shave, then shower.
But in all seriousness, it sounds like you're doing everything right... the only difference being I use soapy water rather than salt water. I also like to secure coffee filters to the tops of my jars with an elastic band for extra protection...
I think the only way we can help is with pictures of your plates and pictures of your grains before inoculation.
When are you noticing contamination? Immediately after inoculation? Half way through?
Even pictures of your contaminated jars may be able to help us.. just need some pics man

Why are you waiting to have a shit and a shave before you do SAB work? You must be bunged up and hairy as hell the rest of the time You don't mention flame sterilization of instruments syringes scalpels etc.

There is a lot of missing info here. First things first, how is your agar work? Clean transfers or do you still see satellites?

Always flame sterilize scalpels and syringes. sorry like I said in tired probably should wait until later too post.
But as far as I can tell my transfers are clean. Once transferred from agar to grain I have great rizo growth. Definitely no satellites.
I usually contam after a few days which leads me to believe its just spores and not just mold. Still waiting them out seeing what happens.
And shower shit shave is just an expression just means cleaning up.

If your agar is clean then your procedure is good. Vector is probably grain, unless you are using poo or coffee etc and pasteurizing incorrectly. I imagine your spawn is bacterial leading to mold when you spawn it.
Best way to test that is to sterilize a quart of grain and leave it for a month. Then put the uninoculated grains to agar. If nothing grows you might be okay.
Other way to test is to do cased grain methods like Vtek or Kramer cakes alongside your spawn jars. If the spawn jars trich out once spawned but the Vtek or Kramer cakes produce, you know the spawn is bacterial.

No trich other things I will update pictures tomorrow. I also have one quart pced. As a variable currently. So far it looks good nothing inside on rye. Most just the inoculated with agar. Playing that waiting game currently I assume its spores because of the germination time on it once inoculated (5-+ days) My agar has hopped to grains at this point.
Sorry for the confusion no sleep sucks
