Agar colonization question updated with pics

Hey guys 5 of my pasty plates colonized and from the looks no contams. I'm wondering what percentage of colonization I should wait for to isolate a strain? (of fast rhizo growth) anybody got a pic so I know what it looks like ?
My plates aren't growing from the center more points popped up since this morning all the plates look like these 3 so where do I go from here ?

You can transfer earlier. If you see sectors cut out the sector you like.

That's true. You can try to take a multispore fruit and clone it. Sometimes it is already an isolate (rare) but there are much less sectors present compared to MS->agar.

Same question here:
I have four agar plates colonizing from a spore print. Since now there's no contamination I can see.
The mycelium grows at the points of inoculation, but I can only see it when I hold the agar plate towards a lamp. It doesn't even look nearly that "dense" like on the picture.
I think I still have to wait some days before transferring to another plate, right?
Or is transferring not necessary, since I can't see any contamination yet?
Do I only have to transfer if I want to isolate or I have contamns, or shall I always transfer?

You should wait some days. At the beginning of growth mycelium is almost invisible. You can transfer to make backups of your ms culture(s).

Of youre I read it, even before I bought my first agar
I am reading a lot, sometimes weeks before starting the entire tek, but sometimes the Teks seem to be described for people who already have some experience, a noob doesn't understand everything. Furthermore my english is school-tought, so urban language is sometimes quite hard for me to understand.
In the mentioned thread he's on cleaning from dishes with contamns.
But my dishes don't have contamns yet, maybe there won't be any, maybe they'll come. I don't know yet.
But I always read: Never let your mycelium colonize too much of the agar, always transfer to other dishes.
But I ask myself: If it's not for isolation purpose, not for a backup and there are not contamns, why transferring and risking to get a contamn? Can't I just wait for the petri dish to fully colonize and if theres still no contamn, directly inoculate grain jars with a piece of agar or tiger drop?

so what your saying is I should make a few fresh plates with these agar to agar for a backup then do a ms grow clone some fruits to agar that leading to a sooner isolate?can I use some wedges to inoculate grains

Of course you can use wedges to inoculate. The problem with cloned fruits is senescence. But you should not worry about that as a newcomer. A lot of people have great success with clones or isolates of clones.

OK I have no contams as of now its been almost 6 days since innoc I'm gonna let colonize then do an agar to grain grow make some clones from a minimono and try to isolate them thank you

You're welcome! If you work with agar you should be as fast and as clean as possible. Keep us posted. You could start a new grow log thread after you inoculated your grain jars. Good luck

Just remember, just because your myc looks clean, contams can still be hiding within it. It's safest to make multiple transfers and monitor.

if you let the plates you have now grow out and they are clean to the best of your knowledge, do like enlightenment says. Wedge or tiger drop, grow,clone and use those plates to possibly isolate. If you wedge and keep the plate you wedged from, chances are it'll pin and you can clone that pin as well. It's a little faster than a full grow to clone.

This is the way to really getting good isolated genetics. I find that a ms tub generally gives at least one awesome clone, and senescence is not usually a problem for quite some time with that clone. I will usually get the best out of five clone candidates, put a tissue sample to agar (from a torn apart stipe, and run my blade down the middle to scratch off an appreciable sample). Ill take a couple plates, grow them, and transfer each sector into another plate. By visual attrition I usually end up with 1 or 2 -great- fruiters this way that are always pretty damned potent.
