So I think I was a little overly eager with my first plates. I think I thought I'd have spore germination quicker. Could be that my measuring spoon for my potato flakes was a tablespoon instead of a teaspoon, and the fact that I almost a tablespoon of agave nectar too...anyway, it's been about ten days, and on the two plates I haven't really looked at at all, I can now see something exactly where I swiped my swab on my plates. The other plate even has 3 lines in a very rough "z" pattern that I tried. Is this what I'm hoping for? See that streak about an inch long?



Also I'd like to note that the condensation on my plates has went down atleast halfway since they were made about 2 weeks ago. Maybe put in the OP that the condensation and frustration of not being able to see does go away when it's actually time to start needing to see.

Assuming Long Term Storage? 

And if so, then yes.  I use pasty plates to store all of my cultures i want to save for later use.  I just pour them a little thicker than normal, let the growth reach the edge, then put them in a ziploc in the fridge.

I don't think it even matters then. I'm not looking thru the side when I take transfers, the lid is off and I'm looking directly at it thru the sab.




But when deciding which plates to transfer from before opening and helping to see any contam present looking through the sides is essential.

I can barely see through the blue lid and thus having to look through the walls with condensation has altered my judgement as to use a plate because I couldn't see well enough to be sure it was clean.

I usually only use the best of the best plates and don't mess with anything showing any signs of weirdness.

I have found a soulution to my visibility problems with no pours and that is these containers...


Other than petris and pour agar this is the best for visibility and condensation. and very durable.

I highly recommend these to no pour users if they have access to them. They seem to be very pricey online but if you have a big lots near you check their tupperware section and look for these containers but the brand is stay fresh.

I think they are $1 a three pack.

sorry pasty I know this tek is based around easily accessible materials but I hope someone reading may have them accessible I hope you'll try them out.

I think I like these moreso for convenience and the really see-through plastic and the clear lid of course. also gives more room for the culture to grow out.

Is the main reason for keeping them upside down the condensation?

I have found, when making transfers, them being upside is much easier as I can leave the lid in place and pick up the container to make a transfer rather than having to remove the lid set on somethething and then pick up the container to make a transfer. I've never been able to cut agar correctly without holding the container.

Thanks to your videos munch I was able to remove a huge vector there. THANKS!!@

I wouldn't say essential. Opening a contaminated plate in a sab isn't much of a risk. I don't think I've ever seen condensation so bad that I couldn't see satellite colonies, thou. Even with condensation you can tell if the growth is uniform or not.

Hey use what works for you. Personally I don't have problems seeing through the sides and I like the small size and easy handling of the mini rounds. But people need to do what's comfortable for them.

I leave foil or bags on jars or dishes so I can put them in the SAB and I sometimes give another spray of water into the SAB. I have a pressurised garden sprayer that gives a fine mist. This way I am not wetting any filters with water from the sprayer.

Slow going... hit 3 plates I think 3 days ago with KSSS spore print. Don't see shit except the black dot of spores...

Maybe soon though :smile:

Also I leave mine in foil, put in SAB, spray soapy water inside one more time then wait ten mins and get Started

I just knocked up 4 plates with burma. The ins9de had a little condensation is that going to be an issue I was on a time restraint and couldn't wait to dry them.

I have left some of mine tilted, so the water gathers at one side of the plate and the rest is dry. If you know they are very likely to have excess water you can let them set in a tilted state, this means you can then leave them on a level surface but the agar will be set in a tilted state and the water will gather in that corner. I have a few jars which are several months old which have filters but still have water in them, it does not always dry out or be reabsorbed as easily as many make out.
