Hey team, I inoculated my first agar plates today, was alot of fun working with my SAB. I'm researching as much as my brain can capitalize and I thought of a neat idea that would be too hard to ask in the search bar

What is better to do? Keep cutting out tiny pieces of mycelium and putting them into new agar plates, or transfering all my colonized agar plates into grain jars and then doing G2G transfers from there to basically have an endless supply of mycelium growing?(not endless, but for my purposes, pretty much)

All I know is I am super excited for this agar tek, this is WAY better than using up entire syringes on a handful of pf cakes so far lol

Isolating a strain from plate to plate for better genetics not quantity of spawn.
G2G for quantity and self replication.

Last link in my signature.

You transfer to new plates to cut down on the amount of strains you have growing together(strain = resultant of two spores mating)

You can take petri dish to a grain jar.

You can G2G your jars to make even more jars

I don't know how to explain it better than if mycelium is on agar you can keep it on agar or do stuff with it on agar and if mycelium is on grain youre on your way to growing.

You're basically asking if tires are better than brakes. You need both things

Ah, gotcha! Agar to agar is to achieve the perfect, healthiest spawn. and of course, if there is bacteria growing, you can isolate the healthy part of the myc, transfer and basically recover from a bad plate right? Are first agar plates seldom perfect?

First agar plates shouldn't be assumed clean. There may be un-germinated contamination you can bring along so its best to take a transfer from some of the fresh growth to ensure you're working with complete cleanliness

Your first plate it made from spores or clone tissue.

So say one rogue contaminate is there. Mycelium is already growing but the contamination is still in spore form so the mycelium grows out on the plate. The wedge you move to plate two can be assumed to be free of any hitchhiker's

I read in another thread, once you got a nice, fully colonized plate, you cut them into quarters, and then each quarter cut into 3 pieces or so, and that is moved to a grain jar, assuming its a 1 quart WBS jar, does this hold true?

Why wouldn't it hold true?
You can throw any sized wedge you would like into a grain jar. Even a whole plate

It would be best to figure out how many grain jars you need to inoculate before cutting up your plate right?
