Has anyone tried stacking these in Qt jars with a lid so they can fit more while having a higher water level in the PC ?

Nope. Looks pretty damn blank. All my other plates I inoculate are still blank...

Maybe I just spazzed out on the one bacterial plate. Hopefully that's the case.

Could just be a dirty swab or print. It happens, that's why we use agar in the first place. Give em a bit the spores might still germ and be able to to be cleaned up.

maybe for you, not for me & my setup though. I spoke of the timings before, the attentive time for me is minimal as I am confident enough to leave the microwave on when I am not there. I can leave my house after 2-3mins work leaving the microwave on for another 20-30mins or so.

I apologize in advance for all the questions. So far I've had inconsistent success with this. Fortunately, just recently, I purchased a proper pressure cooker (presto 16qt) and am now using a sterlite container as a SAB. Before I was using an electric pressure cooker, and a cardboard box as a SAB. Decided to 'upgrade' due to frustration. I'm sure some of my questions have already been answered in this thread but there's alot to read through and thought it might be easier to just ask.

1) After pressure cooking the agar plate there is still some water sitting on top of the plate. Even after leaving the plate alone in the SAB for some time it never solidifies. What did I do wrong? I brought the pressure up slowly (had the fire on low and then gradually increased over time). Although, admittedly, I was a bit impatient so my idea of 'slowly' isn't enough.

2) For some other plates I would have a little water sitting on top. But then it would solidify after sitting the sab for some time. I believe this is normal/okay/satisfactory?

3) The aluminum foil covering the PP5 container would turn black/burned/charred at the bottom (the part touching the water). Is this something to worry about? Did I overcook? Whenever the PC goes a little over 15 PSI I would just adjust the heat to get the needle as close to 15 PSI as possible.

4) Any recommendations for a good scalpel/exacto knife? The scalpel I bought from amazon couldn't be flame sterilized (only the tip goes orange hot). It was a #11 blade I believe.

Damn I had no idea agar could be this simplified.. The couple teks I peeped were pretty intimidating to the average noob but this seems real beginner friendly.. Thanks for taking the time to share this..

1)  try this
22) Potato dextrose yeast agar: 10g agar, 10g potato starch, 7g dextrose, 1g yeast


2)  You can pour  the water out in the SAB by opening the lid a tiny amunt upside down

3) Standard

4) any metal scalpel that you can flame will work.  If you flame it and it doesnt turn red, it doesnt mean it isnt sterile.  Its the heat of the torch that burns all the contamination of the blade.  If you put any metal blade under intense heat for 5-10 seconds its sterile.

The microwave heats too unevenly for my taste. I have never had any luck with it.

it doesn't need to get completely red. aslong as it sizzles when cooled then you're fine

I realize a PC is ideal; but how might this be adapted to boiling (or microwaving) sterilization?

Boiling or microwaving is not an option. It is possible with a high contam rate but who wants this? Ebay -> pressure cooker.
It is worth the money.

agar can be sterilized without pressure, steam sterilize agar for 60m at least. also do not use grain soak water or home made PDA. use "lab grade" media that dissolves completely leaving a clear transparent solution with no sediment or turbidity/opaque. it's not ideal but you can get away steam sterilizing some media that way.

Alright guys, I'm a bit worried here. I just pressure cooked some new plates but I don't think I put enough water in here. I'm using the presto 16-quart cooker and put 1.5 quarts of water in there (which is the minimum). Whenever I put more the pasty plates float in the water and aren't still. When I was done pressure cooking for 45 minutes there was still a little bit of water left (the water level was under the rack).

1) Do the pasty plates need to be in direct contact with water for the entire duration of the 45 minutes to be properly sterilized/cooked? I suspect it was only properly cooked for 20 or so minutes before too much water getting evaporated. Leaving the plates getting cooked above water.

2) Was I supposed to be 2 or more quarts of water in there? Won't that increase the chances of the plates tipping over and being cooked on its sides?

I'm panicking a bit here. I won't be able to use the pressure cooker again until Thursday. My roommates HATE the sound it makes when it's on 15 psi.

Use jar rings to elevate them if the rack isn't high enough, you don't want them in contact with the water. A few inches is enough, I only use an inch worth myself. It should be plenty for the run.
