Chlorophyllum rhacodes aka Shaggy Parasol, clone attempt.
    #23745725 - 10/17/16 02:10 PM (7 years, 3 months ago)

So I recently found several good sized patches of a wonderful edible species called Shaggy Parasol:
https://www.shroomery.org/forums/showflat.php/Number/23740148
The ID was confirmed by a TI.

I took 6 stipe cuttings to agar and so far at least 2 have shown new growth!
There is a fair amount of bacteria but I'm confident I can clean the cultures up.

This species is supposedly culturable by standard methods because it's saprobic.

I'll update this post if anything comes of this.
I hope to do several indoor and outdoor grows with this species, as well as a culture giveaway...

Update, 4 plates out of 7 displayed obvious growth from the clone tissue.
Only 2 were free of any contaminants AND showed growth.

Here they are!

This species bruises brown/red so heavily that it actually dyed the agar brown on some of the plates...

Nice!  Would you grow them on leaf litter or compost of sorts?

They were growing near a row of windbreak pines (and one large maple tree).
Heavy leaf and pine duff + dirt.

I will probably use a mix of leaf litter, aged pine needles and compost.

I am going to try some CVG and manure grows indoors too.

Update:
One of the plates got a colony of bacteria near the edge of the dish, probably from mishandling of the plate during inoculation.
I would usually just toss it, but since I don't have much of this culture I decided to cut the original transfer from the plate and divide it to two new plates.

They fuzzed up right after transfer and look pretty healthy.
So that makes 3 plates total now.

Still growing very slowly.

Got any white paper? Bottom left looks like it has a dark ring around it.
Top middle also looks like there may be a dark bit to the left.

They definitely do have a dark ring around the tissue samples!
Good eye.

Reason for the coloration is this species brusies red/dark brown if cut.
The bruising is so strong it actually leaches out in to the agar.

The reason the bottom left has more brown to it is because it's still on the original transfer plate, the others have been moved to new agar.

Pretty neat. Surprised me at first.

I found the old specimen of this species I collected almost 600 feet from the current patch.
Left is the old one, right is the new one.

I collected the old one about 2 years ago.

The top one sure has nice growth, though.  Have you tried different recipes to see if it responds better to varied nutrient sources? i.e. MEA, DFA, etc...just curious...I know you know your shit.

Interesting move Greg, I follow. I have noticed myself, I get similar results with Gelatin.
Gelatin, tea and water has the same effect as agar, starch, tea and water.

Would need to side-by-side, but its interesting.

I found a wild polypore just out of the pin stage, it was growing from a recently dead lodgepole pine.
Took it to 6 plates of agar. (4 gent+MEA, 2 MEA)
Unfortunately I couldn't break it open to get at clean tissue because the thing was so damn dense!
I ended up just cutting bits with a scalpel, it wasn't easy.
It smells like perfume and mushrooms, it's bizarre.

This is now a C. rhacodes and a wild polypore clone log...

The Shaggy Parasol plates have slowed to almost a halt, definitely need something they aren't getting from plain MEA.
I'm going to pop 'em in the fridge and work on them at a later date...
Might try Ferather's tea stuff, peptone agar, etc...

The polypore is showing lots of growth already.
Plenty of bacteria too but it's clean enough to get a good culture:

What is the "gent" mean in gent+MEA agar?
Also, what kind of polypore is that? That brown bruising could be ganoderma applanatum, are you in the Pacific North West of NA? Is the top kind of a pale, dusty brown?

gent = gentamicin = autoclavable antibiotic

No idea what polypore it is, far too young to tell for sure.
Only reason I know it's a polypore is it had already developed a small pore surface on one side.
It smells like perfume and was found on a lodgepole pine.
Slight brown bruising.
I'm in Montana, temps here are dipping below freezing every few nights.

P.S. If anyone wants a sample of this culture PM me.

So I'm glad I didn't put the parasol clones in the fridge yet.
Turns out the myc just started growing much more thinly so I didn't see the new growth, rather than completely stalling out as I thought it did originally.
It's thickening up again too!

The polypore is on it's first set of transfers now, still growing quickly.
Mold colonies on the original clone plates have begun sporulating so it's fortunate I got the transfers out before that happened.

I am very happy to see you got a clean transfer running.

That mycelium is very strong looking.

Probably came from the outside of the fruit I cloned, it was living on a rotting pine tree after all
Doubt I would have got mold if I could have ripped the fruit open to get tissue but it was too dense for that, so I had to cut tissue from the outside of it.

Hmmm, I am worried that colouring is bacteria. It is to far from the mycelium, and far from uniform around it.
Each plate has different patching and there is an obvious difference in colour too.

Or is it really that bad, like a Reishi grow full of bio waste.

I will post micrographs soon.

I've seen that before actually.
You have to consider where he took that clone, presumably somewhere in Rhode Island which is across the states from me.
Also consider the post right below that picture.
Quote:

Terry, that looks very good, when I've tried to grow this, it was never as symmetrical as yours, and the agar stained dark brown pretty quickly.

Here is 1000x, myc taken from near the center of one of the plates.

Not saying it's not possible for it to be contaminated, it just really doesn't look that way to me.

Couple other pictures.
Agar from the area that's stained brown but doesn't have myc on it:

Agar from a blank petri:

Ignore the stains in the same spots on both photos, some random shit got on the camera adapter lens and I'm not sure how to clean it off...

Edit:
Lastly, here is a bit of agar from a particularly slimy plate:

Image one has a love heart, around it is a rainbow pool. Like petrol (gasoline) on water.
Image 2 is clear of said formation, indeed it looks chemical and not bacterial.

Although, image one the 3 green dots have the same pools.

After comparing all three images fully enhanced, the pooling although weird is chemical.
Thats as far as I can go, the reasoning for it is beyond me at the moment.

You have to consider where he took that clone, presumably somewhere in Rhode Island which is across the states from me.
Also consider the post right below that picture.

I enhanced your first two images and uploaded to Imgur: Here.

Did you try pulling-up a concordial bitmap algorithm and just apply it to the horizontal pixels?
