I'm going to make 4 plates today with 3 different nutes: malt extract, BRF, and potato flakes w/ a drop of Karo. I'm super excited to use those little jelly jars. Thanks for the awesome tek, Pasty.

What ratio do you use for your Grainwater Agar? I might try out RGS next, depending on how my research goes, and I like using grainwater agar every now and then as a starch source. usually do 100mls GW for every 500mls agar

Jelly (preserve) jars are the tits, pity I am struggling to find replacement lids for mine,PP5 plastic lids would be great if they fitted well and sealed properly.

How do you get dry ingredients for a gw agar puck? Should I gently evaporate my water in an oven and scrape up the results?

3 EZLC. 2 TYC agar wedges nocced up 1/2. 1 PE Biopsy nocced up 12/31. They are a little smaller than a quarter. Do you think I should wait or add sterilized water now?

Hell ya. This is my second LC ever I tried another method but the brf was so damn cloudy I couldn’t see shit. With a little more visibility hopefully I can have someone with good eyes tell me what’s up.

Does this look right? It's growing off the puck, but not spreading/clouding up. The color is red food coloring

hey pasty, when i go to suck up the lc, is it safe to stick the heated needle while red hot into the rtv port? i want to limit the contam risk as much as possible

I'm new to mycology and interested in trying this to create a LC to inoculate bags but I have a few questions before I start.
Am I safe with PCing my jars of sterile water at the same time as the agar jars and letting them sit while the agar cultures?
My lids have SFDs for gas exchange on the exterior. Will I be ok or do these jars get shaken much?
Will 18 gauge needles be an adequate size for aspirating the final LC into the syringe?
If for some reason my technique sucks and I introduce contam when adding the sterile water to the agar jar, will it show up visibly as the LC grows in the water?
Thanks, I appreciate any guidance.

Yes PC your water in advance. Won't hurt.
SFD will be okay, I tend to just "swirl" the colony rather than shaking the fuck outta it. That will keep the filter from getting soaked.
18g is pretty small. 16g would be better IMO. This method produces a very thick LC.
If your procedure is poor you may contam the colony when adding the water. I suggest you test the LC before using, especially if you are a novice.

Sorry if this has been asked, I've been drinking and keep spacing out. I need to ask before I forget and I wanna try this soon.
You said you're loose with the starches and sugars, but when you make up your agar do you add (we'll go with for this example) malt extract calculated to your final LC concentration or just strong agar?
Basically what I'm asking is if I want 100 ml of LC at 3% nutrients, should I add 3 grams of malt extract to the little bit of agar or should I go a little lower?

I would say you could add the 3% to the agar. I usually am looking to make a super nutrient puck with this and while the myc won't run as fast on the puck with the extra nutes we don't really need it to. It will take off once you add the water anyways.
