what is DFA? it's never a bad Idea to change up nutrition sources. I would say pour a kinda thick puck. next time I plan on using a regular mouth quart on it's side to make transferring to it easier.

I personally prefer to change nute sources. As for a quart of LC you could use a thicker puck and a more nutritious media. A quart of LC is a fair amount.

I would just pour it myself. In fact I mostly pour and just use syringes to aspirate these days.

So a quick question about Whatman filters, i looked at the whatman site and found 116 different syringe filters, um which one do we like to use?

What's the purpose of the SHIP in the lid? I have some of the blue ones and just got 60cc syringes with 18 guage needles and whatman filters. Do you solely use it to inject water once colonized to size of a quarter, then only use the filter to aspirate into syringe for usage?

Don't use the filter to aspirate. That's what the SHIP is for. You can use it to inject/aspirate if you are less confident in your ability to pour.

no pour would be best with those. they are just a little awkward to handle in the SAB

It took some more reading but I understand now; the whatman filter isn't actually used to filter the syringe, it's mainly for gas exchange while the SHIP does the injecting and aspirating. I wasn't looking to do pouring anyway as I already have tried that with BLI and while the results are decent I'd like something with quicker growth.
I prefer pouring agar over no pour but for LC this seems very simple

even with pouring it should be faster than bli because of the extra nutes and myc growing in there. the main reason I say no pour would be best is because you need to sterilize the jars anyway. petri dishes come sterile. I don't think jars do

If the whatman filter is for GE why couldn't you just do like a 1/8 inch hole with MP over the top? Just curious as I'm sitting here waiting for the silicone to set so I can throw them in the PC

Micro pore is a poor filter for liquids. But if you read the tek again you will see my alternative options to whatman.

some filters can't get wet. period. the filter keeps the jar from forming a vacuum seal and pressurizing the vessel when you extract the water
