Halting Trich growth in colonizing grain jars with peroxide?
    #23740655 - 10/15/16 07:00 PM (7 years, 3 months ago)

I read in a book a while ago called "Growing Wild Mushrooms" by Bob Harris that he puts a few drops of peroxide into his uncolonized grain jars before doing g2g transfers. A little over a week ago I inoculated 7 quart jars of rye that had been sterilized for about 75 mins at 10-15 psi. 5 jars were PES Amazonian and the other 2 were from a treasure coast syringe. Both spore syringes mere MS and acquired from a vendor on facebook for about $5 a piece. The TC jars contaminated after only a few days, while the Amazonian showed healthy mycelia growth. The contaminated jars were removed immediately from the closet shelf which I have my jars colonizing on. A few days later I noticed upon very careful examination tiny tiny green spots in my Amazonian jars. There is maybe only 1-2 tiny mild spots in each jar and none is larger than maybe half a grain of rye. The ratio of colonized mycelium to mold at this point is about 30:1 and I've read that the mycelium can often win out, but I'm still worried. My question is if a sterile syringe filled with a 5-10% peroxide solution could be used to squirt a little peroxide in each jar to help combat the tiny mold growths. I know that peroxide will kill mushroom spores but it should be safe for live mycelium, which is present in the jar in several spots. I would include pics but the mold specks are so tiny it's almost as if they're not there until you look suuuuper close. Has anyone ever tried injecting peroxide to save colonizing jars from Trich infection? It may be important to mention that my jar lids are constructed with 2 holes punches in them, one plugged up with polyfill for FAE and the other sealed with RTV silicone for self healing injection ports.

I would say if it is definitely trich, then they are doomed; maybe wait it out to be sure. As for the peroxide, it will weaken live mycelium, not good. I will refer to this post for the proper use of H202. https://www.shroomery.org/forums/showflat.php/Number/23738247/fpart/1/vc/1/nt/2

Adding h2o2 isn't going to do anything to help with mold. Not at all.

Your jars are toast. They're no good for g2g or spawning. Chalk it up to a learning experience.

Take your spars to agar next time so you can clean them up.

Thanks for the replies guys! Yeah I figured my jars were pretty much screwed and the h2o2 really only works for misting a colonized substrate or cleaning up a workspace but figured Id still ask. As soon as I noticed the mold I inoculated 7 more quarts of rye, 2 with the remaining ms syringe of treasure coast that I had and another ms spire syringe of golden teacher from the same vendor, so even if these jars succumb to Trich (I'm betting they will) I at least have 7 more to keep my fingers crossed on. So my next question here would be what did I do wrong? I've successfully grown with pf tek before and got 0% contamination, yielded about 60g dry of PES Hawaiian from 10 cakes. This was from a syringe I made myself from a sporeprint made by a friend.

My method for inoculation is thus: once rye jars have been sterilized for about 80-90 mins at 10-15 psi (seems to fluctuate as I cook), I let them cool overnight in my cooker. Sometimes I will take them out to set them on a shelf but this shouldn't matter as I leave foil on top of lids until inoculation. I have a small 4x5 storage area that I use for inoculation. About 1 hr prior to working, I turn off the air (room has no vents but I like to be extra safe). I then remove everything from the room besides the TV tray I use as a work table. After putting on gloves I sanitize the air with Lysol, wipe all surfaces down with alcohol and then spray more Lysol. I then sterilize my glovebox by wiping with alcohol in the sterile environment and then I wipe my jars, spore syringes and lighter with alcohol before placing in the glove box. Glove box is sealed and then I completely douse the room in Lysol for about 120 seconds, making sure to get every corner and also the ceiling. Usually it becomes very hard to breathe due to the amount of Lysol I use so I'm sure that I use enough. Afterwards I usually smoke a bowl or two and let the room settle for 30 mins or so. After that I wash my hands and put on clean clothes, I then spray myself with Lysol from head to toe before entering the room. I then put on gloves which I sterilize with alcohol swabs and then wipe down the outer packaging of the alcohol swabs which I use with alcohol before placing them in my glove box. After this I wipe my needle down with alcohol, flame sterilize, wipe with alcohol again to cool the needle and inoculate.

What am I doing wrong here? To my knowledge my technique is sterile and I have had success before using the same techniques with brf jars that were only steam sterilized prior to inoculation! Is the problem with my spore syringes or my technique? I have about 8ccs of oak ridge spores and several other prints that I believe are sterile due to the source. If need be I can test these same results with a syringe that I make myself from one of these prints or with the oak ridge syringe that is already made up, I just want to know whether you guys think it's my technique or just a contaminated syringe that dooomed me from the start?

I know it's kinda long but I wanted to be as detailed as possible, thanks for all the advice!

You made the same mistake I made when I started. Alcohol won't kill mold spores in your glove box. It is more of a sanitizer, not a sterilizer. Use bleach instead. Also ease up on spraying the room with Lysol, no matter how much you spray there will still be mold spores. No need to take a Lysol shower lol. Just clean your tools, jars, hands, arms etc. before putting them in the SAB and eliminate any drafts.

Do your jar lids have filters or just open holes?

Could be a contaminated syringe, maybe try another.

Ive heard from others that a bleach water solution works very well, I'll have to try that with my next inoculation: I will also be making my own syringes from clean prints that I have rather than order from a vendor I had never heard of (got mycelial growth at least so they weren't totally bunk). My jar lids have 2 holes punched in the top, one sealed with rtv silicone and the other jammed with a chunk of polyfil so I don't think my lids are the issue although I'm open to suggestions on how to improve them. When you say "clean" do you mean wiping down tools, arms, hands etc with bleach? Also thanks for saving me from continuing to inhale copious amounts of Lysol while I work lol

I would really like to test out spore syringes and LC on agar but I've never worked with it and at this point idk if I'm experienced enough to actually know whether my spores were bad or if I just fucked up while transferring my spores to agar lol

Thanks for all the responses you guys are the shit!

Finally got some good pics. Mold growth hasn't spread much for the past few days but it appears to be making mycelial growth far less aggressive and I think these jars need to be thrown out in a few days. Always hate wasting spores :/
