Hidden mold/bacteria on petri?

Can mold or bacteria ever not be clearly visible on a seemingly healthy culture? Like everything looks great from the start and you keep transferring til u get an isolate but when u put it to grain it fails.  Is it usually just because its weak mycelium?

unlikely but it can happen. But the biggest risk is the final transfer to grain. When people catch mold spores from the air and put them with the agar into to the grain jars.

I guess I just got to make it a priority to transfer the wedge to the jar as quickly as possible.

its generally pretty easy to ID a bacterial culture as it makes the mycelium to strange things growth-wise. mold can be more difficult, but generally it will just weaken the mycelium, making it appear thin. when it reaches 100% colonization, there is visibly less mycelium than with a healthy jar.
