Proper procedure to inject a myco bag.

I'm trying to hunt down a contam issue with my Golden teacher/petri/LI syringe/presealed Myco bag/ franks monos

I saw this this morning, greenness near my taped injection hole. and I shook/mixed the bag after injection.



I wipe with alcohol, masking tape, wipe again, inject, and very quickly tape over that. making sure all adhesive sticks well.

I don't understand what you're doing with that masking tape, you tape up your tape or something? I would also not wipe an exposed injection hole with alcohol as that can introduce contaminants. What I do is wipe with iso, flame needle, inject, tape up hole. I also do not mix my bags up until they have some visible growth because if there is some co tamination in there mixing it will give it more of a chance of overtaking the mycilium.

Whether my procedure is totally sterile I don't know, normally I will always get contams after awhile bc I use a lc and eventually something gets introduced into it with so many injections, but for the first few sets of bags I create I do not tend to have any issues using this technique. This is all done in front of a flow hood. If I did not have a flow hood I wouldn't bother even trying bags as doing them in the open air is like playing the lottery.

That all was great to hear.

A long time ago I read a tek on making spore syringes with a ziplock bag. It worked great for me, and that guy said to tape first, to keep from tearing the plastic. I'll change that.

I do the whole thing in open air. I'll change that.

I mix it immediately. I'll change that.

Oh I see what you're saying about the pre tape. If you do that I'd put that tape on before pressure cooking. You could also put some masking tape on and then use some silicone caulk to create a self healing injection port which will survive the pressure cooker. If you don't use the tape it won't stick to the bags well.

The bulk bag grains need to be lots drier than jar ones.

I prep my grains the same either way. You can probably get away with more wet grains when there's less maybe but if they're properly hydrated they're properly hydrated.

How fine is the line for to wet or to dry for bags with 4-5 quarts or quart jars?

I'm not sure how to really quantify that. I know I've under or over hydrated grains a lot through the years though, eventually you literally just get a good feel for it. I also use rye these days which is a lot more consistent. Previous to that I would use wbs which not only varies per bag/brand but the grains themselves being all different sizes and with differing starch content makes it more difficult to nail down.

I remember RR stating once somewhere that properly prepared grains can't be too dry... The water inside them after being cooked is enough. I've taken his word on it and I have a huge screen door I spill 16 quarts of rye on for about 10 minutes with a pillar of steam. They feel about right after that. Quite dry.

I prep half a 5 gallon bucket of grains at a time. I'll fill them in one of those mesh paint strainers made for 5 gallon buckets and set them outside to dry. Take an hour or two depending on the environment but it's the best way I've found to process bulk grains like that.

The canning rack inside the canning pot you buy. I set a screen in that and it makes a wonderful huge strainer. Fits everything I boil in the pot it comes with.
