For my first agar transfer (from MS syrgine), the growth was not entirely circular.. why?

It looks like two organisms are growing. The one part on the bottom that's like a half circle reminded me of fluffy tomentose mycelium. The other part looked more rigid and flat and almost like it was possibly not even cubensis. And the outer ring looks really cool, is that mycelium? What do you think?

In any case, I didn't want them to keep growing. I put a sample of each corner into a new plate to isolate them to see what happens.

Its only going to be a perfect circle when its an isolate. One drop of spore solution contains lots of spores and they are all partnering up and growing side by side. The growth contains lot of separate growths with different genetics.

You need to do an agar sandwich on tht hoe.
That will trapp any bacteria contamination beneath the agar sandwich.  Only the clean mycelium will pull through.  As soon as you see mycelium growing from the surface scrap some off and transfer to a new dish.  Repeat if necessary until your left with a clean culture.

No cube mycelium grew that was sourced from that plate :frown:  A couple of them developed a spikey/rigid "mycelium fingers" type look. And all of them smelled like yeast or bacteria.
