Thanks
10 samples were taken. Its been about 24 hours and they are all showing signs of growth. Might make a new SAB just for agar work.
The mix is coming along, but nothing worth photographing. The bald spots are closing up.
Thinking tomorrow morning it will be completely covered. Do you switch to fruiting conditions right away, or give it a few days?
Once this one is given fruiting conditions starting the next one!

dope! good job buddy, cant wait to see your progress in a few weeks
i think you got pretty lucky in many respects so far (nice rhizo culture, fast colonization, more organized than lots of ms). hopefully, you wont ever have to inoculate with straight MS from a spore syringe ever again if you keep learnin and stay on course
no consolidation time required for monotubs, just introduce fruiting conditions as soon as it is 100% colonized. make sure it is 100% for sure, since less is asking for contamination and uneven pinsets
leave out your old agar plates in the light, and after they colonize 100% they will often pin, and then you can clone a pin from agar and go straight to clone culture without having to fruit MS. alternatively, clone a pin from a nice cluster in your current tub, and do a transfer or 2 to make sure it is clean and organized, then make grainspawn from that
btw here are 2 SABs i built, including my current one:

On Thu Oct 20 2016 02:09 PM, c10h12n2o said: sorry to hear that my friend!! dont be embarrassed, it happens to all of us, especially when we first start. if you think critically, you can learn WAAAAAAAAAY more from your mistakes than from your successes
people might be mean or flame you, but a lot of the time you can learn even from those comments, if you dont let them bother you. get what you came for, and fuck all the noise: you came to learn. the good part about sharing (though it takes some serious humility/perspective to not care about all the jackasses) is that there are hundreds of lurkers who dont post but will read your thread and learn from your mistakes and successes, and even from the mean bs. i do it for the lurkers, because i have always been one lol...
do you have any pics?
people always get really attached to their first tubs/jars/bags/whatever, on a sentimental level haha... but i advise as soon as you see green mold, bag it up and throw it away, as far away from your projects as possible. I do it outside in my garden, i actually get my gf(basically wife) to do it so i dont get trich spores on me, and give her a spray bottle of 10:1 bleach to spray it down with, and she showers as soon as she gets in and is extra careful with handling the clothes
trichaderma agressivum is no joke, it will ruin all your projects if you let it. does bread mold quickly in your house? if so you will have to be extra careful. that shit is so fast, so aggressive, and it is parasitic to cubensis cultures. people often have to bleach bomb their whole grow area if it gets established too well. i hate that shit
btw, did it start from those uncolonized patches? or where did it originate?
focus on your projects that are likely to succeed and learning from this one. your cleaned up, organized cultures will be tons of fun to work with
you could bury it outside and it might fruit, ive seen that work a suprising number of times. but i would spray it down with bleach and trash it, unless you have a good spot and climate for that
you just used CVG right? how did you pasteurize? people have success with bucket tek but i would never do that, i use proper pasteurization for everything. Pick up a "roaster oven" on craigslist for $20 (ive got 3 this way) and it is perfect for pasteurizing 3 gallon bags of sub at a time. your problems probably came from bad pasteurization, or unclean spawn. any uncolonized grain or partially sterilized (too hot while pasteurizing) or unpasteurized portion of substrate will get trich QUICK in open air. also, if you can, test the PH of your sub (not essential, just optional), since an acidic sub is prone to trich, might need to buffer it a bit
what size are these tubs? best standard is 66qt. some people like mini monos, i like big ones. i use buckets for my mini stuff

btw, if you do decide to post this in your thread, please include my message as well, for the benefit of any lurkers who find themselves in the same situation
monotubs are about the simplest, easiest, most reliable way to do bulk, and big ones are easier to maintain microclimate in than small ones. what i love about them is you can set and forget them like i did in college (fruiting at spawn, polyfill from the beginning, left in a friends closet, at a storage rental place, and in a dorm; left for 4-6 weeks, came back and harvested, total neglect tek), or you can baby the hell out of them, with the lid off during fruiting, supplying as much FAE and water as possible and keeping constant evaporation. Definitely master the monotub thing if you want to grow cubes bulk just dont get discouraged, it would be surprising if your first tub didnt contam. yoo have actually been really lucky so far, i think you have some relatively aggressive genetics, which helps give your culture the edge in becoming established before contams have a chance. you are going to shit a golden monkey when you harvest your first flush
with that first one in the trash, tell me about what else you have going. 10 pints? of what, and how did you make it? is it from the same exact culture as the other, or have you done a few transfers and then went agar to grain? if you can get an aggressive, organized culture on agar, transfer it to grain and you are ready to rock
id do another monotub (after we work out what needs to be changed about your approach), using all 10 pints of spawn, and about the same amount of substrate as spawn, ratio of 1:1 or 1:1.5. alternatively, you could do a monotub with 8 pints and a little mini otto bucket like mine with the other 2 pints
warm regards

I would post this on my thread, but Im too embarrassed.
A few days after stuffing the holes and there is a good amount of green mold . today there are ~15 pinky nail sized blotches of green almost all of which is located near the central area of the tub.
Im at a loss as too how it happened regardless Im wondering if it should just be scrapped or if fruiting is still possible.
My untrained eyes are unable to tell how close it is to fruiting, and I see no pins well maybe one forming in the corner but thats it.
Truly bummed out about it possibly ending like this, but In some ways Im thankful that this happened now to this small test batch then if I had gone through with the full scale one first.
The next one is raising concerns now too. I was thinking just putting it all in a massive tub, but that would mean if there was another contamination the whole thing might be wasted, have ~10 pints of spawn ready to go. maybe should just make lots of small chambers. idk.
Thanks for helping me out. its been a rough few days. Dr_S
Your right I can take a bit of humility to better the community, and myself. Take note people! This is for all the lurkers out there!
So as you may have gathered the grow is overrun with the sticky icky green stuff.
Here are some pics It all started with this, a weird looking slightly green discoloration. It wasn't a spot that was slow to colonize, nor was it close to one of those spots.
Here is the tub, hurts to look at. and a close up. its actually turning from green to white in some spots.

Here are some of the stretchyer photos of the jars. slight yellowing discoloration, not sure if this is due to the nature of the rye berries getting older or something more sinister. The jars had also been shaken a good amount as they colonized.
jars have been fully colonized for almost a week and though it looks wet now the top half was actually quite dry before moving it around for examination. wanted to hold off until I figured out whats going on before using them.
How deep would you bury it? next week will be in the mid 50s with lows of high 30s is it even worth trying it with those temps?
I attempted to pasteurize it in jars, though I think it was too hot. <---- this was a bad mistake
Also the black liner might not have been sterilized well enough.
Used CVG though ended up using a fair amount more verm then coir, the coir didn't expand as much as advertised.
roaster oven thats a good idea
At least the agar samples are going well. no contaminants. Most of them are about half way to the edge of the dishes (about 3in diameter dishes). Think my agar is a little lame on the nutrients, the growth is not very thick.
Have ~10 pints colonized rye berries which were g2g transferred from the original MS spawn.
actually was going to use a 90qt tub its ~2 ft2 Im down to get a smaller one if you think this is too big. Also id like to try a bucket or two as well. yours produced great results! Doing some maths now I found 8 pint would NOT be enough for a 1:1 ratio at 3 in for the 90qt tub.
For those of you who like maths... volume needed for 90qt tub 1ft(Width) X 2ft(lenght) X 3in(hight) = .5ft3 8pt(spawn) = .13ft3(volume) That would make for a 1:3 ratio .13 ft3(spawn) to .39 ft3(CVG). So to find the perfect size .13 ft3 X 2 = .27 ft3(1:1 volume) then .27 ft3 = 3 in X L ft X W ft -> 1.1 ft2 = L ft X W ft
Im looking for about a 1 ft X 1 ft tub for 8 pts spawn @ 1:1
Thanks for the encouragement C10H12N2O oh how that monkey will sing when the day comes!

So its looking a lot like the failure to properly pasteurize the substrate may be the underlying issue.
Ill be starting a new thread soon for my next go at it. Ill post a link at a future date for those interested.
