How many isolates from a multi spore or clone

Hi, Just a quick 'How do you do it' question.
So how many isolates do you go for.
Also once you have isolated how do you store them?

From what I've read: more of the vets and TC's go for a single isolate, but MANY of them. They test for speed, potency and such. They pick the best ones and clone.The same people are using slants for storage and g2g or LC to continue lineage. They may also use agar wedges from the isolate to inoculate more agar to continue lineage as well.
There may be other methods used but they will have to tell you "their" preferred method.
Again.....just from what I read.

Not many create isolates from multispore, it's a lot of work and then all the isolates have to be tested. What most people will do is do some multispore grows, take some clones then test the clones. The best ones are then slanted on master slants and stored in the fridge. Much faster and more reliable this way.

Im determined to grow me an isolate. I always get close and then get bored with the nagging and stop.

Ive seen someone say to pour some extra thick plates and grow an invitro fruit in one. Clone said fruit. Sounds like a great idea to me. I havent tried it but common sense states it would produce a strain that can fruit in not the best of conditions and will pin fast.

Yeah I did this the last time as I was being lazy and let it go too long
On that subject is it still ok to use a plate that is growing pins invitro for Liquid Inoculant

but that wouldn't exactly get you any closer to isolating a strain if you are doing this from spores.

Search for roger rabbits tek. It definitely works. He shows it on his video series.
Plus more than likely if the pin came from a MSG it isn't necessarily an isolate. It's a good way to start though.

That would be an invitro clone, not a true isolate. But yes, many people do this.
