Okay, so I have some agar glad minis in my PC right now, and my cakes fruit are really starting to take off. I'm going to be taking a portion of the nicest cluster, fastest growing mushrooms to try and isolate via agar. My question is: What part of the mushroom will I be scraping into my plate of agar? I've read somewhere to cut the mushroom in half and scrape from the inner portion of the stem, this seems to be my likely route. Any tips on that part? Also, as I'm planning to go bulk with my agar, when should I prepare my WBS jars in terms of how long it will take to work with the agar growth? I've been reading and searching but I'm not clear on either of these parts

Dont cut the mushroom. Tear it apart. I always take from the base but I only do that because I read it in TMC a long time ago. Works for me. You will really benefit from watching a video with someone doing it. Plenty on youtube including a bootleg of the best (RRs.) Its not so easy if you havent worked with agar before in a SAB. It took me a couple tries to get a rhythm and I wish I would have seen RRs video sooner.
How long its on agar is up to you. If that fruit came from a multispore germination I believe I have heard somewhere that a fruit isn't necessarily an isolate. Someone check me on that. So you still have some isolating you might want to do. Also if a plate contams (the first one probably will) you have to keep transferring until the plate is clean before you can use it. Dont sterilize grains until the night before you want to use them. When a plate is satisfactory to you, do your grains that night and xfer in the morning.

Thank you so much, I totally didn't even think to watch a video.. I'll probably spend an hour or two doing that tonight. They did come from a ms syringe, and I'm hoping to try and isolate Malabar Coast fr it because I'm in love with how it looks. Money and time isn't an issue (thank god for legal drug dealing... Er, bartending hehe) so I'm prepared to spend a while working with this agar and getting as much practice and experience. You've really helped me put this into scope, I was feeling a little clueless. I had all the pieces of the puzzle in my brain but you just helped me put it all together.
