Tarragon Oyster give away
#23680797 - 09/26/16 06:42 PM (7 years, 4 months ago) |
|

Ferather was kind enough to send me a culture of tarragon oyster growing on dowel pins from the UK. Well they finally arrived in great shape. He specifically asked to have Tarragon distributed in the USA for people to enjoy. Thank you Ferather!
Please visit Ferather's grow thread to learn more: https://www.shroomery.org/forums/showflat.php/Number/23611616
Keeping to my word I plan on making limited batches of agar wedges, master slants and possibly LC syringes to give away to people who wish to give this interesting fungi a whirl. The only thing I ask is to cover shipping. More information will come later when I get closer to sending out the cultures.
For now some work needs to be done. I have to put these to agar and do any cleaning if necessary. Im hoping none will be needed because the high quality dowels I recieved. I will chronicle my work in this thread to keep everyone updated.
So here we go
Day 1:

I was going to put these to agar tonight but decided to wait till tomorrow to give the myc some time to adjust and rest.
---------------------------------------------------------------------------
Day 3:
I put some of the mycelium from the plugs on agar to get an idea on how clean the culture is. Unfortunatly as I suspected there is other stuff growing with the myc especially trich and possible bacteria. However I have one plate that is showing the beginning signs of a different mycelium growth structure. Hopefully there will be some oyster mycelium for me to get a sample of. The plate that has turned green is being tossed but the ones that still have white mixed growth I will be keeping till either I have the opertunity to isolate oyster mycelium or just toss them.
Tonight I plan on plating more mycelium and a wood plug to agar and starting a very small batch of grain spawn to see if I cant get a section of good oyster mycelium growth.
On a good note the summer oyster plugs that Ferather sent as well seem to be pretty damn clean. I have started to see good mycelium growth with no contaminations yet. He did a damn good job on those plugs!
Here are some pics of the first Terragon Oyster agar attempt:

Some of you are going to notice the little dots running away from the mass of mycelium. Those are from me tapping the agar with the forceps to release the mycelium. It isnt bacteria but it is the start of trich.
---------------------------------------------------------------------------
Day 6:
It has been a very busy day. Started alot more spawn, made some agar plates, started new LC jars, made some slants and isolated Terragon and summer oyster. It's crazy how fast time can blow by.

Here is the update on the Terragon oyster. I had to toss a few more agar plates due to trich being the primary growth. I do however have a very promising plate that is showing good oyster mycelium growth with only two small trich spots and the oyster mycelium seems to be over growing the trich.

From this good plate i made four more new plates from the good new mycelium growth. Hopefully at least one of these will end up good so I can continue to isolate.

Also on the 29th I added some mycelium and two plugs to cardboard to see if a good growth point could be taken. I just noticed today that I have good starting growth and I have yet to see anything that resembles trich but it is too early to celebrate. I have my fingers crossed though.

Im really hoping to have a clean culture or close to having one in a couple weeks. If any one has any suggestions that might add a different method for success please let me know.
---------------------------------------------------------------------------
Day 13:
Sorry this update is running late. I have been out of state for work. We were put on call for Hurricane Matthew. Thankfully Im home now and didn't get activated.
I was able to finally get clean mycelium from the Tarragon Oyster using a SAB. Keep your fingers crossed that no spores from the trich got onto the isolations.
Here are some pics.

I hope these isolations take and within a week or so I can take the final isolation onto antibiotic agar. Im still on schedule despite the trich issue. So guys and gals say some prayers to keep the trich at bay.
------------------------------------------
Day 17:
Just got home from working out of town for the week. As soon as I got in the door I ran to my lab area to check on the Tarragon Oyster cultures. Totally expecting a Trich infected mess I was pleasantly surprised to see the start of two clean cultures. No sugns of bacteria or other competing fungi. YESSS!!
So very very soon I will have cultures to send out to people.

---------------------------------------------------------------------------
Day 26
Here is the latest update. I have clean mycelium growth. Im at the final stage and I can start sending out agar wedges to people who want them. I plan on starting master slants and some LC solution today.

Thank you katbusa and Ferafather. This is very generous of you.
I'm still working on mastering my oyster tech but otherwise would jump all over this.
Thanks again for giving back to the community.

I'm more than willing to ship to Canada. When I ship up north I watch the weather and use heater packs if needed. Should have no problems.

Thanx mate, I should have included Canada in the conditions. A really bad move that I fully apologize for.

Thats annoying, So TravelAgency was correct with dowels not being sterilized for outdoor use. The zip bag I cleaned with antibacterial soap and ethyl + isopropyl alcohol.
Both continue to work for 30 minutes after rinsing.
Try a disc of cardboard, grow out and cut. Then transfer to agar, might work.

Don't sweat it Ferather. I wasn`t expecting it to be easy. I was thinking last night and Im going to give the cardboard a shot and hydrate it with your antibacterial tea tek. Im crossing my fingers the type of trich on those would rather eat starches instead of fiber.
I have plenty left to work with so Im going to keep at it till I have a clean culture.

There is a high chance that it is starch loving trich, they look clean in the bag. I can say the vendor dowels where inoculated by a grain sample or two.
I can also add oyster should dominate weak trich. The tea will hold and cut back bacteria.
Should work, in theory.
----
In addition, I got 0% trich from my spawn, if its there it got dominated and failed. So as said, likely to be a starch trich or trich is too slow to adapt.
----
Good luck, Ferather.

I am willing to accept four (4) orders from Canada. Same terms as the original offer.
To those who I have already traded, I can pass on vendor spawn. This offer is only open to those who already traded.
Short delay due to order collection. PM me for further details.

If you are willing, purchase the required materials to make a T-Tek. Its antibacterial and the only competitor is other mycelium.
Make some dishes, you can pull a sample from it. Alternatively, enriched tea slurry agar?

I'm sorry to hear that. It took me prob in the vicinity of 15 plates to get one that had an area of clean myc that I could grab. The trich was a major problem. I was honestly not expecting to get something from these dowels.

Im in the final stage and over the next two weeks I plan on senind out a mixture of agar wedges, slants and lc syringes to people who want them.
I will be sending out agar wedges this week.
Im still trying to figure out how to handle shipping. From experience I would like to send these out VIA USPS priority. The reason behind this is that the shipping time from coast to coast is 3 days and I have had really good success shipping plants and orchids this method with low incidences of damage. It would help out alot if shipping was covered by those who want in on the give away. The average shipping cost would be $6 to $7. Does this sound reasonable?
I hate to do this but I can only ship to USA, Canada and Mexico. I will have cultures available for overseas at a later date.
Please let me know what you all think. Once we get the shipping method hammered out I will post what I have available to ship.

Im more than willing to send you anything you want. In fact you are first on the list. Let me know what you want and I will put it in the mail for you.
The main thing I did was start a bunch of plates. I almost went through two packs of 20 plates. 90% of the plates had heavy trich contamination and were tossed into the garbage. The ones that were promising I kept on the cool side to stunt the trich growth. Then I carefully took samples of good mycelium in a SAB and crossed my fingers in hopes of success. So literally my success was due to lots of samples and using tea as the antibacteria agent in the agar.
In fact the bacterial contamination was totally controlled using the tea method you have been developing. That is why the agar in the last picture set is so dark. Im not trying to prop you up and scratch your back. In all honesty the tea in the agar is what stopped the bacteria that was also in the culture you sent me. It stopped the bacteria so well that I see no need to replate to antibiotic agar. Good research and good science is exactly what you are doing with your tea tek.
I also want to extend an offer for people who want to try Gentramicin Sulfate. Im offering 1 gram of my USP lab grade Gentramicin cut up into 20 50mg servings. Each person will recieve two 50mg vials which is enough to do 1 liter of agar or two 500ml batches of agar. I have enough to cover ten people at no charge. The gentramicin I use comes from caisson labs and you can search the CAS number to see test and purity results on their website.

My first attempt almost, but the vendor spawn was contaminated. It's exactly like Golden oyster. Except no yellow and tarragon (aniseed) smell and tarragon with mild vanilla flavor.
Mine produced tubers, slightly smaller in size than GO tubers.
GO and TO are variations of Branched oyster.

Turns out the Shroomery data is correct and TO has a higher biological efficiency then GO. It appears to have a higher adaption rate and yield potential given time.
Suggested pH range is 6 - 8, 7 is optimal for growth.

Ferather said: My first attempt almost, but the vendor spawn was contaminated. It's exactly like Golden oyster. Except no yellow and tarragon (aniseed) smell and tarragon with mild vanilla flavor.
Mine produced tubers, slightly smaller in size than GO tubers.
GO and TO are variations of Branched oyster.

How was the flavor.. Any yeild comparison to other oysters?
