So with my mono giving me some fruits i will take some to agar and i also have a print ill be doing as well, I do have a few questions if anybody doesnt mind answering

Is potato flakes, malt extract and agar ok for a recipe?

I have a pyrex media bottle do i need to loosen the top and put foil on top to p.c?

And finally about how much should i p.c. to pour 80 dishes?

I think its potato or malt with dextrose and agar. My media bottle has a cap rated to 140°C so foil ksnt necessary if yours has the same

PDA = potato dextrose agar. MEA = malt extract agar, no dextrose added at all.

They're ingredients for 2 different recipes.

25 ml per plate. 25*80=2000 mL (2 litres)

Personally I don't like PDA. I may try making it with reduced sweet potato broth, but I don't like the growth I get on traditional PDA. Dog food and malt extract are my favorite agar nutes.

Thanks for the clarification so is dextrose needed in the pda? Or can i do just potato flakes and agar?

Dextrose is very much needed for PDA. But yeah I too prefer just malt + agar. So easy. 2% malt and 2% agar. Done.

Why would you losen the lid? I did about 600ml didnt fill to 1000ml but it is designed for this purpose id imagine so I put lid on as tight as it would go and chucked it through a pc cycle works brilliant

Actually 40 grams of agar and 40 grams of malt. This is just assuming it has a density similar to water, which is highly unlikely. Idk the exact weight 2% is because I don't know the density's, but I always just remember it as 2g of each per 100 ml. Which is 2% lol

I was getting issues with literally no growth for 3 batches of pasty plates before I realised I hadn't added dextrose...I felt stupid when I realised

I just combine the agar mix and water in the bottle, the pc will homogenize it.

Waiting for the bottle to cool some more but if someone could answer if this is correct

Im going to wait for the bottle to cool a bit more to be able to handle then i will pour dishes and store in bags until cool, inoculate , then wrap with parafilm?

Trying to get freshly poured dishes back into their sleeve would be a nightmare.
Let them cool/solidify in stacks first before putting them back in the sleeve. I usually do stacks of 8.

Truth be told I don't even put them back in the sleeve and I've never had issues (as long as the dishes stay in the SAB).

If you plan to put them in the fridge then you should put em back in the sleeve for sure.

Out of the hundreds I've poured only one has contaminated before I used it.

One other thing, make sure to cut the parafilm in half lengthwise, you don't need the full width to cover a petri:

I don't see why not.
I cut mine like this outside of the SAB.
2 squares (4 in.) length goes around a regular petri once, 4 squares goes around twice and is what I use.


Sometimes I wrap the plates outside the SAB too, but you have to be careful not to lift the lids at all.
