Hello!!!
I just inoculated four Pasty plates with a ms syringe for my first time ever and after 72 hours I'm not sure what I've grown?
Can you identify the stuff below?
Mycelium ..or What???
Plates A, B, C all look about the same, however plate D looks distinctly different:

• Should I try to clean and isolate anything here? Or is it all bad?
• If mycelium, why does it look so exceptionally non rhizomorphic poor?
• If it should all be discarded, what went wrong??
I had tried to search the forum for old posts to see if I could find like examples of growth to identify what I have here, but couldn't.
Bonus Question: How are you supposed to take photos of growth in Pasty plates without taking the cover off? - related: how bad is it to take the lid off for a few seconds to get a better look, so long as it's inside a SAB and you're minding similar sterilization precautions as during inoculation?

Although I've only done agar for a couple of weeks, I've seen a lot of cube mycelium in my life... And it does not look anything like what you've got afaik.
I'm no expert in identifying contams tho, so that's all i can say. I would throw it away.

Bacterial to me I'd bin and start again Pics in pasty plate aren't as easy to get a good pic as in petri dishes imo. Taking the lid off to do so wouldn't be advised unless sterile conditions.

I've just started agar but I'm sure that is bacteria and not mycelium.

Thanks for the feedback, disappointing to hear.
Because there is uniformity of this contamination across the plates I'm inclined to think perhaps it's not my sterile technique, but maybe the agar recipe itself?
..Or could this be how a bad syringe manifests itself??
I used Pasty's agar recipe that includes potato flakes and Karo - but I've seen on a couple other threads now people recommending not including these things, or much in the way of nutrients into the agar itself as this apparently can cause bacterial growth and also cause weak mycelial growth?

In terms of sterile technique: I used a PC, SAB, flame sterilized syringe, showered, alcohol soaked nitrile gloves, breath mask, was careful not to wave hand over open plate, used an empty spare bedroom.
Thoughts? specifically on how I go about preventing this from happening again?

I don't think the agar recipe alone could cause any contamination. If you sterilized the plates well, there are two possibilities. Either there was contamination in the syringe or you introduced contamination as you were inoculating.

In looking at those pics -- given how there's just one type of organism showing in all of those dishes (and I agree now, yes, that looks like bacteria), and how there's no observable presence of mycelial growth -- I think this further isolates the issue to a bad syringe, and not poor sterile technique. No?
I mean, typically if the contamination was brought on through poor sterile technique wouldn't that usually manifest through there being something like a little bit of mycelial growth present but with other contaminants present and taking it over?
