Spores take longer than a day usually. They'll need to grow out more so you can actually separate growth from the original spores germination point This is the peu I took a transfer from at this size which was a little later than I needed to leave it
 This would have been the better time to take it but didn't have time to do it

Hey WillHill,
There is NO WAY those are cubensis spores germinating on that plate. When you tapped out the "spores" on the plate you were inoculating it with everything else that was on the foil, in the bag, etc. You should chuck all that shit and start over with known clean spores or tissue. It will save you a lot of time, resources and frustration.

Thank you for your incredibly direct comments, it's what I really needed to hear, I really am just wasting my time here.
I have ordered up a fresh spore print and a loop, is there anything else I should get in preparation for a fresh start?
When spores do arrive, any advice for getting them from the foil to the agar?

Do all work in SAB follow sterile techniques iso, gloves, flame loop, spray SAB soapy water etc. Literally touch the print with the loop then swipe/dab it on plate. Be careful but quick, I'm not saying move fast I'm saying have your movements planned and everything arranged in SAB where it needs to be its a good idea to do a dry run just so you know where things need to be so your not scrabbling around with the petri lid off trying to grab something you put behind your arm or something. That's what I do anyway

It will be at least a week maybe 2 before they get here, I'm going to use this time to build a low cost HEPA filter and fan setup.
I made a quick solid works drawing..

You bet! Flame your loop till glowing red. You shouldn't need to cool it in the agar as loops cool off pretty quickly...then gently drag the loop across a small section of the print and then gently drag your loop back across the agar. All this should be done in a still air box or in front of a hepa filter of course. Definitely follow up on sterile technique and so on as mentioned above. You'll be very happy with your results if you do some real research on agar, technique, etc. and apply what you've studied. Your spores will germinate around 5-7 days, typically. Good luck!

If it works well and is affordable it would be very interesting to see the main thing that's put me off is the cost of a flow hood but I'd love to have one

This I've designed at 50cm wide x 40cm deep for the footprint on the desk, i dont have much space to give it more and including the 99.9% filter, fan, germicidal bulb, regular strip light and plexi glass housing and a few buttons I'll give it a budget of 200 dollars.
Should be a decent challenge, the hardest bit will be mounting the fan and impeller on a bearing. I'll order up the plexi now.

I got the case laser cut today and then drilled and tapped the screw holes.
12mm thick acrylic, lets see how easily it scratches over time, I may swap the table out for stainless steel one
I'm confident that the 316 stainless M5x30 screws will never rust.

Just waiting on the other parts now

Why do we need to separate growth from the spore? What happens if we dont?

Thanks for that, I just saw them in allot of commercial flow hoods, I guess the idea is to turn it on before use for a while to kill anything on the inside of the hood. But as its redundant ill not bother adding one.
