How risky is a full plate?
    #23671732 - 09/23/16 05:36 PM (7 years, 4 months ago)

Hey all

I always thought you shouldnt łet ýour culture fill the entire plate- at least not disposable Pitres.  leave  room if its going into cold storage and let the culture grow out the rest of the way. Its nice to see if its still clean.

But aside from cold storage, and of course not letting the myc climb out the walls, is there any signifigant risk to letting the plate fully colonize?

I imagine it's less of an issue for PP5 plates with high walls like Pasty Plates?
Thats actually why Im asking is because Im innoculating grain masters but I left my cultures out a little too long and the myc is reaching up the wall some.... and I  was thinkjng Hmm.. maybe I should transfer once more to be safe.

This is something I never had a definitive answer on. Figured what the he'll might as well just ask

The reason you don't want them to grow to the edge is in case some satellites are starting there. You want to be able to see them. If a mold colony is covered by the mycelium it will grow through the agar into the grain.

If you are concerned then just cut a 1/2" from the edge of the plate. Only major issue is you are loosing the most vigorous part of the colony at that point but I doubt in practice that it will hurt that much.
