I need help with a contamination problem.
    #23659755 - 09/19/16 06:59 PM (7 years, 4 months ago)

Franks Monos with Magnetic swappable filters | Petri dishes with numerous transfers | Pre-sealed Myco Bags nocced with blender made LI with syringe | coco/verm substrate with improved bucket tek | Golden teacher | rye berries | misting/fanning irregularly

Hi,

I've been doing monotubs for years, and shotgun jars before that. Still a novice though, I know my place.

So I asking for some help.

I am getting contams after my first harvest, most of the time. I'm only getting 3 ounces dry from a tub with over 3 quarts of rye grains. My stumps are often rotting. Then mold. (see attached photos (hours after a dunk)

My ideas for the possible cause:

1) Someone smart in the forum recently spoke of the possibility that bacteria is fighting mycelia during the grain consolidation, making it weak, but not visibly so - causing it to be prone to disease.

2) My bucket tek (temps measured and timed, then cooled quickly).... is inadequate. Need to pasteurize my sub better.

3) My myco bags suck.

4) Something about the environment I'm providing is less than desirable (sub moisture, FAE, misting/fanning, spawn/sub mixing cleanliness)

Hit me with your gut instincts. I want to fix this. Thanks in advance.

Your spawn is the culprit.

My spawn prep:

Agar dishes, numerous transfers... clean looking to me.

Blender made LI

3.5 quarts of rye in a bag, presealed. four bags in a pressure cooker, careful wooden spacers to circulate steam. 3.5 hours.

Alcohol spot on bag, tape that, more alcohol, flame the needle, inject 15 mm of LI. quickly tape hole. shake.

It takes over the bag, but not real fast. I have been seeing growth starting in 6 days or so. Perhaps 2.5 weeks to be complete.

I recently learned that my practice of cooling my agar petri dished before making the LI weakens the mycelia, even if I let it recover at room temp for a day. I'm changing that now.

I am very fond of pre-sealing the bag, because all I have is a crummy Still Air Box, and love seeing the bag all sealed when it comes out of the pressure cooker.

crummy SAB?  a shit one is awesome. dont hate

id just transfer wedges of agar. LI sounds cool but its another step that
can introduce contams..

also if it goes sour after 1 flush your spawn is fucked.

I have a still air box

OK. I'd like to see if I can fix my current procedure before I change things up too much.

Is LI injected into a myco bag a really bad idea?

I get creeped out bringing an unsealed myco bag out of a PC, opening it in a loving beautuful SAB, then dragging it over to a sealer. Doesn't sound so cool in my not so clean apartment.

I would guess that if your plates really are clean, the problem is somewhere in the process of making the LI.

Maybe I'll try a squirt and scrape in the petri dish method.... it has worked well in the past.

Another thing about that open bag thing in the PC.... doesn't the PC, when cooling, suck a bunch of room air into the chamber?

You can also give GLC a shot. I don't do bags but I know plenty of people PC unsealed bags.

I'd guess either the bags or the LI. I would try a simpler method for the LI. I fucked an entire round of jars up tying the blender method just due to poor technique. I now do blender less LI. Works better with less vectors. Just do in and shake the agar with water. I'd avoid GLC as it is contam prone.

mushroom tissue will bruise from even the slightest of pressure so your cuts are what is causing the blue, NOT MOLD.

Unless there is more that your not showing...

Mold that I have seen on stumps(actually never seen a blue mold in mush cult) is always green and doesn't show that fast.

It looks pretty healthy maybe slightly bacterial but just keep an eye on it and you should be good,

I totally agree.

OP, keep us updated with pics in the next couple days so we can be sure...

That's when I would usually see mold if my spawn were crap.

Do you use sterile water and sterile syringe to make the LI?

Top left corner of first pic looks a little off to me.  Kinda grey.

I'd agree that the blue on the stumps is bruising.

Top left corner is fine. Just some spores fell there. First pic right in the middle of the pic tho.. those white bubbly things.. that's going to turn green very soon, in fact it looks like there's already some trich sporulating just to the left of it. Definitely take some close-ups of this area OP.

OK,

There is other mold. Both photos show the beginning of a greyish contam.

But the stumps are rotting/becoming slimey.

This is the first I've heard of this... bruising. I use sharp scissors snips. And when I dunk, I flood the sub with water and tip it up vertical with a gloved hand to drain it out for a minute.

How can I improve?

as said before, this is most likely because of dirty spawn which either had trich in the first place or bacteria which weakened the sub and let the mold set in.

Yeah the slimyness sounds like bacteria...

How long are you dunking for?

maybe cut back a little but that won't solve the problem. You need to focus on find the contam vectors and eliminating them.

Are you sure your agar cultures are clean?(if you are using agar if not start now)

I'm using agar.

I know its toast, thats why I'm here trying to fix it.

So I don't need to change how I'm harvesting, correct?

But I will attempt to get the spawn healthier.

I thought I had it figured out when I learned I wasn't PC'ing my rye long enough. The Big bags need a long ol' time. But that didn't fix it.

Now I'll try removing the blender made LI, make it the old school way in the dish.

I'll post my expanding growth in the bags, to see if any of you can spot ill health.

Many many thanks for your brainstorming, I really appreciate it.

:whathesaid: damn not sure if it's a good thing I can spot unsporulated trich from so far away

Yes up the PC times, ALWAYS have uninoculated control jars to test the lids/filters, and always have clean inoculant. If you do these things and ensure it's fine, you won't have this problem.

Stumps going bacterial and shit is also from bad spawn. They're just as much a part of the mycelial network as anything else. Clean spawn is the key!

Yeah OP if the "stump-rotting and then mold" is what you're depicting in those photos, it just looks like regular ol' bruising to me.

Also, are those exposed grains visible in the first pic?

A day later, one of my tubs has no trich... but the slime is very much no good. This tub looks like I am over misting, I can assure you I am not. Its slimey rot that has weakened the mycelia so much that it looks too wet. Three days after first flush a dunk.

Has it occurred to you to not leave the stumps on the substrate? I actually twist and pull everything tbh lol

There was a conversation between Frank and RR a while back. Frank said he would cut it harvest and get rotting stumps, and get less rot if he twist and pulled. RR brought up that he had contams in his agar stage that weakened his mycelia, and it wasn't the method of harvest that was at the root cause.

I want vibrant, healthy mycelia, and I think my spawn is fucked.

Can this be sterile after being in the pressure cooker after 25 minutes? Its what I've been using to make my LI. I realize its bad... but does rust harbor bacteria?

As a side note, this is what my agar is looking like. This is a quarter slab that fell from my scalpel.. its sharp edged because it was growing up the walls.
