I need help with a contamination problem.

Franks Monos with Magnetic swappable filters | Petri dishes with numerous transfers | Pre-sealed Myco Bags nocced with blender made LI with syringe | coco/verm substrate with improved bucket tek | Golden teacher | rye berries | misting/fanning irregularly
Hi,
I've been doing monotubs for years, and shotgun jars before that. Still a novice though, I know my place.
So I asking for some help.
I am getting contams after my first harvest, most of the time. I'm only getting 3 ounces dry from a tub with over 3 quarts of rye grains. My stumps are often rotting. Then mold. (see attached photos (hours after a dunk)
My ideas for the possible cause:
1) Someone smart in the forum recently spoke of the possibility that bacteria is fighting mycelia during the grain consolidation, making it weak, but not visibly so - causing it to be prone to disease.
2) My bucket tek (temps measured and timed, then cooled quickly).... is inadequate. Need to pasteurize my sub better.
3) My myco bags suck.
4) Something about the environment I'm providing is less than desirable (sub moisture, FAE, misting/fanning, spawn/sub mixing cleanliness)
Hit me with your gut instincts. I want to fix this. Thanks in advance.

Your spawn is the culprit.

My spawn prep:
Agar dishes, numerous transfers... clean looking to me.
Blender made LI
3.5 quarts of rye in a bag, presealed. four bags in a pressure cooker, careful wooden spacers to circulate steam. 3.5 hours.
Alcohol spot on bag, tape that, more alcohol, flame the needle, inject 15 mm of LI. quickly tape hole. shake.
It takes over the bag, but not real fast. I have been seeing growth starting in 6 days or so. Perhaps 2.5 weeks to be complete.
I recently learned that my practice of cooling my agar petri dished before making the LI weakens the mycelia, even if I let it recover at room temp for a day. I'm changing that now.
I am very fond of pre-sealing the bag, because all I have is a crummy Still Air Box, and love seeing the bag all sealed when it comes out of the pressure cooker.

crummy SAB? a shit one is awesome. dont hate
id just transfer wedges of agar. LI sounds cool but its another step that can introduce contams..
also if it goes sour after 1 flush your spawn is fucked.

I have a still air box
OK. I'd like to see if I can fix my current procedure before I change things up too much.
Is LI injected into a myco bag a really bad idea?

I get creeped out bringing an unsealed myco bag out of a PC, opening it in a loving beautuful SAB, then dragging it over to a sealer. Doesn't sound so cool in my not so clean apartment.

I've also wondered about the heartiness of the filter patch to long PC times.

I would guess that if your plates really are clean, the problem is somewhere in the process of making the LI.

Maybe I'll try a squirt and scrape in the petri dish method.... it has worked well in the past.

Another thing about that open bag thing in the PC.... doesn't the PC, when cooling, suck a bunch of room air into the chamber?

You can also give GLC a shot. I don't do bags but I know plenty of people PC unsealed bags.

I'd guess either the bags or the LI. I would try a simpler method for the LI. I fucked an entire round of jars up tying the blender method just due to poor technique. I now do blender less LI. Works better with less vectors. Just do in and shake the agar with water. I'd avoid GLC as it is contam prone.

mushroom tissue will bruise from even the slightest of pressure so your cuts are what is causing the blue, NOT MOLD.
Unless there is more that your not showing...
Mold that I have seen on stumps(actually never seen a blue mold in mush cult) is always green and doesn't show that fast.
It looks pretty healthy maybe slightly bacterial but just keep an eye on it and you should be good,Quote:

I totally agree.
OP, keep us updated with pics in the next couple days so we can be sure...
That's when I would usually see mold if my spawn were crap.
