Assistance in using agar for pf tek
    #23649576 - 09/16/16 05:19 AM (7 years, 6 months ago)

Greetings. I am preparing for my very first cultivation so i would ask you to bear with my ignorance. I have decided to use Pf Tek and a SGFC for my first cultivation however i was also advised to create an agar colony to use for the future. Having purchased a 10ml syringe i will use 8ml for my cakes and the last 2ml on agar. my question is how will i use this agar to create more cakes (cloning or multiply spores) as well as a request for general advice such as where to keep the agar. I appreciate your assistance.

Keep agar in still air box. Second make a lot of plates at same time. You start off with your geration plates. Make a few tranfers at first sign of grow. Keep tranfering until you get clean growth. Same with cloneing pins.
When you have clean cultures you make li. Then inject li like you did spores.
If you can master all that then you should shot for grain spawn.

Use 1 or 2 drops of your syringe....2mls is waaaay too much....once your agar plate germinates..make a couple more transfers to new agar plates and use Munch's blenderless LI to inoculate jars.....that will go further and help with cutting down on contamination greatly...
.you will need a SAB..

I appreciate the advice however i have been told that as my cultivation level will be low (2-3 SGFC aiming for around 100 dried grams every 6 weeks) agar is not necessary. instead i have been advised to simply make spore prints and make new syringes. what are your opinions on this?

Just making syringes will work....but more than likely will be dirty to some point and will cause contaminations eventually....maybe not every time but your success will be greater if you do decide to do agar work...

Even with proper sterilisation and caution? Which part is usually the most dangerous part for contamination when creating spore prints and transfering it into a syringe? I am assuming the spore print itself cannot be contaminated so when loading it up into the syringe?

Acquiring agar itself might be a problem for me. i am unable to find it in the shops and online is not an option. i have heard of home made agar however it just sounds dodgy af.

Your fruits are not grown in a sterile environment therefore your print is not 100% clean, when you suck spores up into a syringe you're loading everything that was stuck to the cap you printed

i understand that the gills might have been contaminated, but how are clean spore syringes created? Im starting off with a spore syringe so are you saying that all spore syringes are contaminated to a degree?

Health food stores carry agar powder too. It's a vegan gelatin substitute and thickener.

I did pf tek 1 time.....ran 6 cakes and got about 21g's dry...do a couple runs in and research going bulk.....IMO it's a lot of work and your dehydrator is gonna be on constantly harvesting trying to dry cakes a few at a time....because they will mature at different times...kinda a pain in the ass but worth it

Yeah PF tek is a good way to do your first grow, but once you get the concepts down bulk is where it's at.
