Liquid culture first time questions
#23646926 - 09/15/16 09:17 AM (7 years, 4 months ago) |

Greetings. I have never grown shrooms before and am trying gain as much knowledge as possible before i start so please bear with me. Firstly, i have seen many mixed opinions about liquid culture in terms of success so my questions is how to maximise success rate and also can you create LC from LC? I thank you for your assistance in advance.

What equipment do you have to start with. Sterility is one of the most important factors

Those silicon injection ports look promising for LC to LC transfers. Have yet to try one out yet though.

If you want to do an LC start with agar, get a clean culture and move from there. Personally I do not and will not use LCs, I make clean agar plates, by the 3rd transfer I usually am confident I have a good clean plate. Then I make grain masters, and from those grain masters i set to work. You can always make fresh cultures or make more masters from a good producing set of jars.

I am not familiar at all with agar plates. Having purchased a 10ml spore syringe i simply want to create a simple system that does not requre much equipment and will give me infinite spores so that i do not have to buy more syringes to inoculate cakes.

I agree with the agar comment whole heartedly
Taking good prints is harder than it sounds. I just recently did my first clean spore syringe. Had to find a PP5 container and toss some aluminum foil squares inside, and then pressure cooked it for like 30 minutes at 15psi and placed into my SAB until I had an good sized cap for printing.

One drop of spore solution onto agar can then be transfered many times to get the cleanest best plate possible and you can take a slice of that plate grow it in grains or make an LC or whatever you want, grow out some shrroms take a print, swab back onto agar start again, with prints you only need a tiny speck of spores onto agar. So a print lasts ages and you can make multiple grows from that speck

I never bother with sterilizing my foil, the cap ain't sterile anyway, just clean up on agar.

Agar is super easy. Take 5 plates, put down a drop on each of them. Transfer and pick the best looking one. Make a master, G2G for 3 expansions. Should take a few weeks. Take the 1000 quarts of spawn you just made and make 200 monotubs. Take 30-50 clones from those, chose the best of them all and have a killer culture to grow forever. Meanwhile throw the 100 lbs of mushrooms you just grew in a bonfire and start growing some more.
Put spore syringe that you used 5 drops out of in a drawer and never look at it again.
Or just fire the syringe into a half gallon of sterile water if you don't want to do agar and just want to stretch the spores. Will still be more than enough.

I always liked to take an inoculation loop and squirt a drop of spore solution on that then inoculate with the loop. Prevents the solution from getting all over your plate.

Instructions un-clear.....filled PP5 with spore solution...
Jk
I also like to swipe the drop with a loop. It kept my growth centered and was more noob friendly.

Then the last thing you need to mess with is liquid culture. Cause LC is used to make 100's of quarts of spawn fast. If you just wanna run a single mini or something LC would be a whole lot of risk for no reason.

I do the same with swabs.
