This is my third hopefully clean agar transfer to isolate, how does it look?

This is 7 days old and it's a pasty plate. I transfer from the leading edge everytime and it's looking more and more uniform with each transfer. How does this look? I started with MS and I'm hoping it's cubensis since I haven't tried fruiting this mycelium yet (and hopefully it's not... oysters or hericium, lols.)

Not bad, but that dark mycelium in the center makes me think that there's a little bacteria under the mycelium. After another transfer or two you should have some super clean myc

Which dark mycelium? You mean at the very center of the original agar piece? Cause I think that's just a shadow. (You could see there's a corresponding shadow on the right side of the plate.)

Did you take that pic inside your sab or in front of the hood? I mean even then i wouldnt take the lid off for a pic unless it was bad already

It was inside the SAB... my apprentice was holding it and i just took the photo with my camera outside the hole and just zoomed it in using the camera. I set the camera to a 3 second timer and took a burst shot (10 shots in one click after the timer sets) so that it would be an ultra steady and clean shot even though it was zoomed in and not the best lighting in the world. Hopefully the picture quality was good enough for you guys to see anything special about it.

Dont worry about it being uniform or not, just look for clean, healthy growth, with no bacteria or mold colonies on the plate. Then your culture is ready to inoculate spawn.
To get an isolated strain/monoculture is quite involved. It usually takes about 8-10 transfers before you even start to see sectoring. Then a transfer from each sector is taken to new plates. Then each of those plates are transfered until they no longer display sectoring and is an isolated strain. Each of those isolates then gets labeled , stored and fruited out to test. Usually only like one out of ten is worth keeping.
Strain isolation is great but is quite involved. I sugguest you use your clean MS culture to inoculate some clean spawn with and maybe find some clones!
