Rye grain inoculation question

Hello there. Recently I knocked my first quart grain jar ever. I am using a jar with a self healing injection port and a filter at the top. My question is this....

I tilted the jar on its side so the syringe would actually push into the grains. I then slowly tilted the jar right side up. After doing some reading I found this may be slowing my colonization time. Its been 4 days and I haven't seen any growth. Should I reinoculate the jar but this time leave it right side up and drop the spores on top of the grain? This would avoid splitting the spores apart. Thanks for any input.

Relax. 4 days is nothing and no. do not re inoculate. Wait a few weeks and see if anything happens

Spores can take a long time to germinate, relax, the magic will happen if your grain is prepped correctly.

If you really want to see the magic, you need agar.

Spores do sometimes take awhile to germinate, but I believe that the length of time it takes it highly dependent on how well your grain was prepped, and conditions.  If it's prepped extremely well, and you're keeping your jars at proper temps, with proper GE and ambient light, it can take 4-5 days.  Sometimes less if you're damn lucky but I've done a lot of MS to grain and generally for me, this is what I expect. 

I've moved to better things since then however.

Thanks for the replys all. The jar is just over 3/4 full. The syringe wont quite reach the grain if the jar is sitting regularly.

You don't need the syringe to reach the grain. Just a few drops on top will do the trick.

Now still no sign of growth but slight condensation at the top of the jar. Is this normal?

Yes.  A little condensation at the top of the jar is pretty normal.  But a bunch around the grain itself isn't.  Causes bacteria to proliferate, and spores have a hard time germinating in such conditions, as previously mentioned.

OK. Temperature is nice. After 12 days in most cases you should see something. I had 4 year old spores that took more than 20 days to germinate on agar. Where did you get your syringe from? From a shroomery sponsor? Is there a 'spore clump' in the middle of the syringe? If there is one: Did you shake the syringe to dissolve the clump before inoculation?

I did indeed shake and i did in fact get the spores from a shroomery sponsor. I bought my rye grain jar from a well trusted site as well. Could it be due to the fact that i tilted the jar and shifted the grains around slightly during inoc? The jar has a self healing port and the syringe wouldnt reach the grain so i tilted the grain towards the injection port. I should have just dripped it on the top. Perhaps i should reinoculate and keep the spores together?

Every drop of water from the syringe should contain enough spores. You dripped in about 1cc that should be more than enough as Pastywhyte already wrote. Everything seems to be OK. I doubt that it has to do with shifting the grains. You could wait this weekend and inoculate again on Monday without moving the jar.

Patience is hard for a newcomer but it is really essential in this hobby.

I wish you luck and success. Don't get disappointed.

21 days later and I see some myc growing where I reinoculated! YESSSS!:grin:

Hi guys, 2 months later and after shaking at 25%. This is my jar....










The average temp is about 63 degrees. It has taken a long time to get this far and im just wondering if the temp is causing slow colonization or the jar has contams. What do you guys think?
