No pour agar: lowest GE for sterilization possible

How small of a hole can I get away with if using an sfd on a no pour such as a pasty plate or similar?

I know agar doesn't really need GE but since its a no pour they would get crushed in the pc unless there is some amount of GE.


How much is is necessary???????

Trying to keep it small to minimize drying out over long periods of time...

I always put a 1/4" hole in the center of my pasty plates and then stuff them tightly with polyfill. Works every time.

I use micro pore tape or tyvek & scilicone. If you use either of these, don't drill in the center, drill on a flat smooth part. It is much easier to get a good seal with the filter. Otherwise you would be filling in all the cracks with scilicone. If you use poly fill it would make no difference.:super:

EDIT: I also use 1/4" hole

I follow the tek 1/4" hole with a couple layers of MP tape, I store new plates in a closed tote. You can also put them in individual ziplocks but they shouldn't dry up for a long time anyway.

I ask because some of my holes got a little small when I melted my filter on and I don't want to ruin the container from too little GE while sterilizing.

Anyone ever used a smaller hole for short term and even lonterm storage?

Seems like not many people use sfd's for agar but maybe someone knows if it would work?

Some of the holes are maybe 1/8" will that slow growth or cause problems during pc cycles?

I know grain needs somewhere around 1/4" for proper growth but what about agar?

anyone know?

If it helps some of mine have like 5 layers of micropore tape over the hole because I just add a new peice each time and pull them off and start over eventually, has to be more restrictive that just a couple but then again the lids loosen after a few runs. I think they'll be fine, test one by itself if you're too worried.

Also mine colonize completely sealed in ziplocks (I know weird) I like that it keeps the outside clean and I don't have to wipe them down later. So the air inside the container is plenty for agar

I use a screw or the tip on my flathead screwdriver on my leatherman I have never had issues but I use micro pore tape
