Alright yeah. I'm entering day 9 here in a few hours and it is small for that point alone. I'm going to sterilize a rye jar right now and inoculate it tomorrow.

Just for the haters who don't believe a simple and sterile GLC to LC can produce a wonderful LC syringe .. or 6. Point being, don't be a dick to someone just because they have a low post count. Everyone is human.

So no fruits at all, hell not even a fully colonized jar?
looks clean at this stage..
 Beginning to have some look bacterial, but had to wait til most of it was colonized..

Lots of bacteria...
Almost all jars look clean at the beginning..

Of course you never know if everything is alright until full colonization... But if the syringe would be contaminated as hell you would see it right now. Chances are high that the jars will colonize just fine.. lets wait. I am sure he keeps us posted...

Here's what a jar covered top to bottom in bacteria looks like. Bacteria is almost impossible to see without mycelium showing its reaction to it. As seen here:

Just saying that there's no way you can say it's clean with barely any myc there. Fully colonized and it's a different story.

You can't see bacteria in LC. Here is some extremely bacterial winecap LC. Can you see the bacteria?

This particular culture took me 4 transfers to clean. It colonizes a jar oh probably halfway (looks completely normal too), before it smells of the black plague and sadness.

All I have to do is zoom in on the 'looks clean at this stage" and I see more than 3 jars with caramel in it. Lol Sorry for being critical on how "clean" they look.

And yes, I will keep this thread updated until the jar is 100% colonized. I live in a very clean town house with zero mold, barely any dust, and I use a SAB with all precautionary measures. Contams don't just form, they enter your "play" area.
Let's see how clean my play area really is.

You mean agar wedges?... Yeah that's common with agar 2 grain . I seriously hope you don't mean the agar wedges lol

Research on endospores. Farmers spray grains with them as a form of natural pesticides. This is the reason why you have to PC grains much longer than you normally would for sterilization, and can easily survive a 3 hour pc cycle. Aloha does a double cycle for 4 hours each to kill endospores (sterilization of most media is less than 20 minutes @ 15 psi). That's why grain LCs are considered bad practice. Can they work? Sure. But you're literally just gambling. It's the same as tossing a coin.

Endospores are easily killed by a PC however protected by grains it becomes difficult also sterilization time depends on initial CFU load.
